摘 要:目的观察气道平滑肌细胞(ASMC)经致敏血清刺激后嗜酸性粒细胞趋化因子(eotaxin)的表达变化及沙丁胺醇对其表达的影响,探讨其可能的作用机制及β2受体激动剂在哮喘治疗中的作用。方法无菌条件下分离SD大鼠气管段,组织块贴壁法体外培养ASMC。建立SD大鼠哮喘模型,采血收集血清。以致敏动物血清刺激体外培养的ASMC并收集培养上清及细胞。采用酶联免疫吸附法测定培养液上清中的eotaxin水平;放射免疫法测定培养细胞胞内环磷酸腺苷(cAMP)的表达;逆转录-聚合酶链反应半定量检测培养细胞中的eotaxin mRNA的表达。结果哮喘大鼠被动致敏后,气道平滑肌细胞可表达eotaxin(n=4,P<0.05),其表达水平与胞内的cAMP水平呈负相关(P<0.01,r=0.788);沙丁胺醇干预后,ASMC中eotaxin的表达明显下降(n=4,P<0.05)。结论气道平滑肌细胞在致敏状态下eotaxin的表达显著增强,作为炎性分泌细胞参与哮喘发生过程,是哮喘发病机制的重要组成部分。沙丁胺醇可以在哮喘发病中有效抑制炎性细胞因子的表达。[ Objective ] To examine expression of the CC chemokine eotaxin in vitro rat ASMC induced with serum from asthmatic rat and observe the subsequent effect by treatment with salbutamol; To investigate the role of ASMC autocrine and its signaling pathway in asthma. [ Methods ] ASM was isolated from rat trachea:chemokine and cAMP expressions in ASMC were examined by radioimmunoassay, ELISA and reverse transcriptase-polymerase chain reaction. [Results] There were significant eotaxin protein and mRNA expressions associated with cAMP level (P 〈0.01, r =0.788), compared with control group in ASMC induced by sensitized rat serum(P 〈0.05), and apparently intervened by sulbutamol therapy (P 〈0.05). [ Conclusions ] There is an autoefine function in ASMC as an inflam- matory cell on sensitized condition as part of the central role in asthma. Salbutamol can depress the expression of inflammatory cytokine effectively.
