摘  要:目的:通过预先椎管注射氟柠檬酸(FCA)抑制脊髓胶质细胞功能后,观察电针足三里穴对调控糖尿病胃轻瘫大鼠胃电活动及延髓内神经元和星形胶质细胞功能的变化,以探讨脊髓胶质细胞在针刺调控胃运动中的作用。方法:50只雄性SD大鼠分为空白对照组(空白组)、糖尿病胃轻瘫模型组(模型组)、模型组+电针足三里穴组(足三里组)、椎管注射FCA+模型组+电针足三里穴组(FCA组)和椎管注射生理盐水+模型组+电针足三里穴组(NS组),每组10只。模型制备采用腹腔注射5%四氧嘧啶和熟地灌胃诱导的方法。实验3周后记录各组动物胃电活动,并对各组动物延髓内fos和GFAP进行免疫组织化学染色。结果:与空白组比较,模型组大鼠的胃电平均频率和振幅明显降低。而足三里组和FCA组的平均频率和振幅较模型组有明显升高(P<0.01);以足三里组升高的更明显,且与FCA组比较,差异有统计学意义(P<0.05)。除空白组,各实验组fos和GFAP阳性表达产物集中表达于延髓迷走孤束复合体(VSC)内,有明显的定位特点。其中以模型组的表达最高;足三里组与模型组比较,阳性fos数量明显减少,阳性GFAP表达明显降低(P<0.01),;而FCA组与足三里组比较,阳性fos数量和阳性GFAP表达均有所增加,且二者之间差异有统计学意义(P<0.05);模型组与NS组之间差异无统计学意义(P>0.05)。结论:椎管注射FCA对针刺调控糖尿病大鼠胃运动功能有抑制作用,脊髓胶质细胞参与了针刺调控胃运动的作用。Objective:To study the function of spinal cord glial cells on acupuncture in regulating the gastric motility,by pretreated with Fluorocitrate(FCA) and electroacupuncture(EA) administration,we observed the changes of the gastric motility activity and the function of neuron and astrocytes in bulbus of diabetic rats.Methods 50 male SD rats were randomized into 5 groups,Control group(Control),diabetic gastoparesis model group(DGP),DGP plus EA Zusanli acupoint group(Zusanli),intrathecal(i.t.) injection with FCA plus Zusanli group(FCA)and(i.t.) with NS plus Zusanli group(NS).DGP model rats was induced by intraperitoneal injection(i.p.) 5% alloxan plus poured libosch into the stomach.After three weeks,the animal's gastric motility activity were recorded.The expression of Fos and glial fibrillary acidic protein(GFAP) immunoreactivity were stained in bulbus.Results The DGP group's average frequency and wave amplitude were greatly reduced,compared with Con group's.When administration with EA Zuanli or(i.t.) with FCA,the average of frequency and wave amplitude were increased;and these changes were bigger in Zuanli groups than in FCA groups.The expression of positive fos and GFAP were significantly increased in the vagal-solitary complex(VSC) of bulbus.Image-analysis demonstrated that there were greatly difference in the DGP group,FCA group and Zuanli group.The DGP group was highest,the FCA group middle,the Zuanli group lowest.There was slightly difference the FCA group and NS group.Conclusion Administration with FCA(i.t.) can retard the function of EA on gastric motility in DGP rats,the spinal cord glial cells may play a role in these EA's effects.
