百草枯中毒对大鼠的肺损伤作用研究

摘　　要：[目的]观察一次性灌胃染毒百草枯对大鼠肺组织的损伤情况。[方法]将20只大鼠随机分为对照组和百草枯组,百草枯组一次性染毒百草枯50 mg/kg体重,对照组用生理盐水代替,观察大鼠一般情况变化,染毒前后对大鼠进行跑步机训练,比较2组大鼠灌胃前以及灌胃后第5、10、15天跑步机上掉落次数。于染毒后第15天麻醉处死大鼠,收集肺组织,碱水解法测定肺组织羟脯氨酸含量,HE染色观察大鼠肺组织病理学变化。[结果]百草枯组大鼠灌胃后一般状况差,百草枯组平均进食量较对照组显著降低。灌胃前大鼠跑步时掉落次数2组间并无显著差异,但灌胃后百草枯组掉落次数较对照组增加。百草枯组大鼠肺组织羟脯氨酸含量极显著较对照组高,病理切片显示染毒组肺组织肺泡腔塌陷,肺泡间隔明显增宽,间质呈纤维性增厚。[结论]百草枯一次性染毒导致大鼠肺组织纤维化。[Objective] To observe the lung injury of rats with once intragastric administration of paraquat.[Method] 20 rats were randomly divided into control group and paraquat group,rats in paraquat group were given 50mg/kg paraquat only once,while that in control group were given saline instead.Then the treadmill training of rats was observed before and after intragastric administration,the times of rats falling from the treadmill in the two groups before and 5,10,15 days after administration were compared.Rats were killed in the 15th days exposure to paraquat,their lung tissues were collected to measure their hydroxyproline content by alkaline hydrolysis and observe the pathological changes by HE staining.[Result] The general condition of rats was poor in paraquat group.Their average food intakes were significantly lower than that of the control group,the times of rats falling from treadmill in those two groups differed insignificantly before administration,but the falling times of paraquat group increased than the control group.The hydroxyproline content in lung tissues of paraquat rat was significantly higher than the control group,pathological section showed alveolar collapse,widened alveolar septum,and fibrous thickening of the interstitial in paraquat group.[Conclusion] One time paraquat exposure would lead to lung fibrosis in rats.
