摘 要:目的探讨3种藏药提取物体外抗腺病毒和甲型H1N1流感病毒的作用。方法采用细胞病变法检测3种药物对Hep-2细胞的毒性作用以及其抗腺病毒的活性作用;采用MTT法检测3种药物对MDCK细胞的毒性作用,并通过蚀斑法观察3种藏药提取物甲型H1N1流感病毒的抑制作用。以治疗指数(TI)值作为药物抗病毒效果的评价指标。结果 3种藏药提取物A、B、C对Hep-2细胞半数毒性浓度分别为0.872、0.872、0.438 g/L,抗腺病毒作用的半数有效浓度分别为0.244、0.288、0.250 g/L,治疗指数分别为3.574、2.819、1.752;3种药物对MDCK细胞半数毒性浓度分别为1.076、1.745、0.399 g/L,抗甲型H1N1流感病毒的半数有效浓度分别为0.240、1.663、0.891 g/L,治疗指数分别为4.491、1.050、0.447。结论 3种藏药提取物具有一定的抗腺病毒和抗甲型H1N1流感病毒的作用,具有较广的抗病毒谱,其抗病毒作用强弱程度依次为A、B、C,说明其具有潜在的临床应用前景,值得进一步深入研究。Objective To investigate the inhibitory effects of 3 Tibetan medicine extractives against adenovirus and influenza virus A(H1N1) in vitro.Methods The cytotoxicity of extractives was evaluated for Hep-2 and MDCK cells.The antiviral effects against AV3 and H1N1 were investigated by the assays of methyl thiazolyl tetrazolium colorimetry(MTT) and plaque reduction.Results The median cytotoxic concentrations(CC_50) of extractives A,B and C were 0.872 g/L,0.872 g/L and 0.438 g/L on Hep-2 cells,and were 1.076 g/L,1.745 g/L and 0.399 g/L on MDCK cells,respectively.The median adenovirus inhibition effective concentrations(EC_50) of extractives A,B and C were 0.244 g/L,0.288 g/L and 0.250 g/L,respectively.The median influenza virus A(H1N1) inhibition EC_50 of extractives A,B and C were 0.240 g/L,1.663 g/L and 0.891 g/L.The treatment indexes of extractives A,B and C were 3.574,2.819 and 1.752 for adenovirus,and were 4.491,1.050 and 0.447 for influenza virus A(H1N1),respectively.Conclusions The extractives of 3 Tibetan medicines had inhibitory effects on adenovirus and influenza virus A(H1N1).Further research needs to focus on the mechanism of the antiviral activity.
