摘 要:目的探讨实时荧光定量逆转录-聚合酶链反应(RT—PCR)法检测富含半胱氨酸的酸性分泌糖蛋白(secreted protein,acidic and rich in cysteine,SPARC)对瘢痕疙瘩成纤维细胞β1转化生长因子(TGF-β1)、I型胶原蛋白基因表达的影响。方法用不同浓度重组人SPARC作用于体外培养的瘢痕疙瘩成纤维细胞,并以空白组为对照,实时荧光定量RT-PCR法检测TGF-βI、I型胶原蛋白mRNA的表达。结果与空白对照组相比,实验组TGF-βI、I型胶原蛋白mRNA表达明显增高(P〈0.05)。结论SPARC能显著促进瘢痕疙瘩成纤维细胞TGF-β1、I型胶原蛋白基因的表达。Objective To observe the effects of secreted protein, acidic and rich in cysteine (SPARC) on the expression of TGF-β1 and collagen type I in cultured human keloid fibroblasts by real-time fluorescence quantitative RT-PCR. Methods In vitro keloid fibroblasts were stimulated by different concentrations of recombinant human SPARC, and with the control group for comparison, real-time fluorescence quantitative RT-PCR to detect expression of TGF-β1 and collagen type I . Results Compared with the control group, the expression of TGF-β1 and collagen type I was significantly increased in the experimental group. Conclusions SPARC could enhance the expression of TGF-β1 and collagen type I in keloid fibroblasts significantly.
