摘 要:目的制备特异性和非特异性猪脾转移因子(Transfer factor,TF),并比较二者的理化特性及免疫活性。方法采集免疫猪和非免疫猪脾脏,采用冻融-透析法提取猪脾特异性和非特异性转移因子,检测二者的理化性质;E-玫瑰花环试验检测二者的活性;利用环磷酰胺复制小鼠免疫抑制模型,MTT法检测二者对小鼠淋巴细胞转化的影响。结果特异性及非特异性猪脾转移因子蛋白含量分别为0.392和0.026 mg/ml,核酸含量分别为1.130 6和0.987 7μg/μl,无菌试验、热原试验和安全试验均未出现异常,符合相关规定要求。特异性和非特异性猪脾转移因子均可增强小鼠腹腔巨噬细胞的吞噬功能,增加E-玫瑰花环的形成率。特异性猪脾转移因子对免疫抑制小鼠、生理盐水组小鼠及正常小鼠外周血淋巴细胞转化均有明显的免疫增强作用,与对照组相比,差异均有统计学意义(P<0.01),且特异性猪脾转移因子的免疫增强作用显著高于PHA组(P<0.01);而非特异性猪脾转移因子对免疫抑制小鼠、生理盐水组小鼠及正常小鼠外周血淋巴细胞转化均无明显的免疫增强作用(P>0.05),且明显低于PHA对照组(P<0.05)。结论特异性猪脾转移因子比非特异性猪脾转移因子具有更明显的免疫增强作用。Objective To prepare specific and non-specific transfer factor(TF) from pig spleen and compare their physiochemical characters and immunological activities.Methods The spleens of immunized and unimmunized pigs were collected,from which TFs were extracted by freeze-thaw-dialysis,and analyzed for physiochemical characters.The activities of the prepared TFs were determined by E-rosette formation test.Mouse model of immunosuppression was copied by using cyclophosphamide,based on which the effect of the TFs on transformation of mouse lymphocytes was evaluated by MTT method.Results The protein contents in the prepared specific and non-specific TFs were 0.392 and 0.026 mg / ml,while the nucleic acid contents were 1.130 6 and 0.987 7 μg / μl,respectively.The TFs were qualified in sterility,pyrogen and safety tests.The TFs enhanced the phagocytic function of mouse celiac macrophages and increased the formation rate of E-rosette.The transformation rates of peripheral blood lymphocytes of immunosuppressive mice,normal mice immunized with physiological saline and unimmunized normal mice after treatment with specific TF were significantly higher than those in control group(P 0.01) and those after treatment with PHA(P 0.01).However,non-specific TF showed no immunopotentiation to the transformation of peripheral blood lymphocytes of immunosuppressive mice,normal mice immunized with physiological saline and unimmunized normal mice(P 0.05),of which the transformation rates were significantly lower than those after treatment with PHA(P 0.05).Conclusion The immunopotentiation of specific TF was stronger than that of non-specific TF.
