摘  要:目的观察大鼠尾吊模拟失重3周及解悬吊恢复8周后骨密度(BMD)及骨力学(biomechanics)的变化情况,探讨五加补骨方对其干预效果。方法雄性Wistar大鼠,按体重随机分为4组:对照组(Con)、模型组(Mod)、五加全程给药组(A),五加恢复期给药组(B)。A组全程给予五加补骨方水煎液灌胃,B组在8周恢复期给予与A组等剂量的五加补骨方水煎液灌胃。3周尾吊结束时及解悬吊8周时,取材测量不同部位BMD及骨力学性能。结果与对照组相比,尾吊3周后模型组、A组和B组的股骨、胫骨、第四腰椎BMD显著下降(P<0.001),股骨最大载荷和弹性载荷也显著下降(P<0.01);A组的BMD下降幅度明显小于模型组和B组。解悬吊8周后,和对照组相比,模型组股骨、胫骨BMD仍未恢复至正常水平(P<0.05);A组和B组大鼠的股骨,胫骨,第四腰椎BMD已恢复至正常水平(P>0.05);模型组,A组和B组股骨和肱骨最大载荷和弹性载荷同对照组比无显著变化。结论尾吊3周模拟失重可以造成大鼠承重骨BMD和骨力学显著降低,而五加补骨方具有明显对抗骨丢失的作用;在恢复期,骨密度及骨力学恢复缓慢,五加补骨方在一定程度上可以促进大鼠骨丢失的恢复。全程给予五加补骨方对抗骨丢失的疗效明显好于解悬吊后再给药。Objective To observe the effects of a Chinese recipe-wujiabugu,on bone mineral density(BMD) and biomechanics of rats after 3 weeks tail suspension(TS) and 8 weeks de-tail-suspension recovery.Methods Fifty-six male Wistar rats were randomly divided into group control(Con),group model(Mod),group Awujiabugu recipe and group Bwujiabugu recipe.During the experiment(11 weeks),group A was given wujiabugu decoction.Group B were given demineralized water during TS,then wujiabugu decoction in reambulation.Following TS and 8 weeks recovery,their BMD and biomechanics were examined respectively.Results At the end of TS,compared with Con,the BMD of femur,tibia and the 4th lumbar vertebra of the other 3 groups significantly decreased(P0.001);The femur maximum load and elastic load decreased(P0.01) too.BMD in Group A was much higher than those in group Mod and B.After 8 weeks recovery,there was no significant difference in BMD and biomechanics of femur,tibia and the 4th lumbar vertebra in group A and group B when compared with group Con(P0.05).BMD in group Mod was still significantly lower in femur and tibia when compared with group Con(P0.05).There was no difference in bone biomechanics of femur and humerus.Conclusion TS for 3 weeks can lead to significant decreases in BMD and bone biomechanics of bearing bone of rats;wujiabugu recipe can relieve weightlessness osteoporosis and save the recovery time of BMD and bone biomechanics in bearing bone.
