摘  要:目的:观察rhIFN-γ对离体小儿瘢痕成纤维细胞生物学作用的影响。方法:分离、培养小儿挛缩瘢痕组织中的成纤维细胞,加入不同浓度rhIFN-γ进行刺激,采用MTT法检测细胞增殖,流式细胞术分析细胞周期,分光光度计测定胶原含量,透明座标纸测定成纤维细胞胶原网架(FPCL)的直径并计算收缩指数。结果:rhIFN-γ达到一定浓度后,成纤维细胞的增殖活性明显下降(P<0.01);细胞分裂受抑制,G1期细胞所占比例增高(P<0.01);胶原合成减少,FPCL收缩指数下调(P<0.01)。结论:rhIFN-γ是成纤维细胞的负性调节因子,能抑制成纤维细胞的增殖分化,下调胶原纤维含量及成纤维细胞胶原网的收缩性,起到抑制瘢痕增生的作用。Objective To observe the biological effect of IFN-γ on the proliferation of in vitro fibroblasts derived from children's scar.Methods Fibroblasts in children's contracted scar were used cultured in cell culture medium containing different concentration of rhIFN-γ.The proliferation of the fibroblasts was measured by MTT assay,the cell division cycle was analyzed by flow cytometry,the quantity of collagen was determined by spectrophotometry and the contractile index was caculated besides measuring the diameter of the fibroblast-populated collagen lattice(FPCL) by clear scale-paper.Results When the rhIFN-γ reached a certain concentration,the proliferation of fibroblasts significantly decreased,and there were significantly differences between control group and the treatment group(P〈0.01),The percentage of G1 phase cells in the treatment group was significantly higher than in the control group,The difference was significant (P〈0.01),the synthesis quantity of collagen and the FPCL contraction index in the treatment group were obviously lower than in the control group (Pall 0.01).Conclusion As a negative adjustment factor,rhIFN-γ,by inhibiting the proliferation and differentiaation of fibroblasts,down-regulating the content of collagen fiber and lowering the contraction of connective tissue,could control the hyperplasia and contracture of children's scar.
