摘 要:目的:探讨人胚胎早期发育中Wnt/β-catenin及NF-κB的表达及意义。方法:采用免疫组织化学SABC法对30例3~12周人胚胎心发育中Wnt1/β-catenin及NF-κB的表达进行检测。结果:胚胎3周时,组成心管的全层细胞呈Wnt1免疫阳性;5周时,心外形已基本建立,心内膜及外膜层细胞呈β-catenin免疫阳性;8周时,心壁增厚,成心肌细胞互相连接成网状,心肌层细胞及心腔内皮细胞呈β-catenin和NF-κB免疫阳性并持续至12周。结论:Wnt/β-catenin及NF-κB因子在人胚心早期发育及心肌细胞分化中起着重要的调控作用。Objective: To explore the expression of Wnt/β-catenin and NF-κB in early development of human heart, and therefore, to understand the significance. Methods: 30 fetus aged from embryonic week 3 to 12 were sampled according to the principle of informed consent, and submitted to examine the expression of Wntl/β-catenin and NF-κB using immunohis tochemical S-ABC method. Results: At embryonic week 3, the all cells in cardiac tube were exhibited positive to Wntl. The immunoreaction of β-catenin was detectable in endocardium and epicardium at embryonic week 5, when the outline of the heart was formed, and were positive against. Additional three weeks later, the wall was thickened and the cardioblasts were reticulation. At the same time, the myocardium and endothelial ceils of the cardiac chambers were found to be positive to β-catenin and NF-κB, and upregulated till embryonic week 12. Conclusion: This finding suggests that Wntl/β-catenin and NF-κB may play an important role in early heart development and cadiocyte differentiation.
