慢性鼻窦炎伴鼻息肉与HLA-DPB1、DQB1基因相关性研究

摘　　要：目的:探讨慢性鼻窦炎伴鼻息肉患者在HLA-DPB1、DQB1两个基因位点的易感因素及保护性因素,初步探讨HLA-DPB1、DQB1两个基因与慢性鼻窦炎伴鼻息肉的相关性,寻找易感基因类型。方法:通过对HLA-DPB1、DQB1两个基因位点进行PCR扩增,用ABI 3700自动测序仪对扩增产物进行测序,从而对46例广东籍慢性鼻窦炎伴鼻息肉患者和50例广东藉健康对照组HLA-DPB1、DQB1两个基因位点进行分型及比较。结果:DPB1*040101等位基因仅出现在病例组;DPB1*0502-030101和DQB1*0202-020101两种等位基因频率在对照组高于病例组,差异有统计学意义;DQB1*050301在病例组中的频率高于在对照组,但差异无统计学意义。结论:DPB1*040101及DQB1*050301可能为慢性鼻窦炎伴鼻息肉发病的易感因素。而DPB1*0502-030101和DQB1*0202-020101在慢性鼻窦炎伴鼻息肉的发病中起保护性作用。Objective：To determine the contribution of the Human Leukocyte Antigen（HLA）-DPB1 and DQB1 genes to Chronic Rhinosinusitis with Nasal Polyps.Methods：A comparative case-control study with 46 patients and 50 controls was performed.HLA-DPB1 and DQB1 loci were typed by polymerase chain reaction（PCR） and the PCR products of the amplification were sequenced directly on an ABI 3700 automated DNA sequencer.Results：HLA-DPB1＊040101 was only found in patient group.The frequency of HLA-DPB1＊0502-030101 and HLA-DQB1＊0202-020101 was lower in patient group than control group.They showed statistically significance.A higher frequency of DQB1＊050301 appeared in patients with Chronic Rhinosinusitis with Nasal Polyps,but the P-value was not statistically significant.Conclusion：HLA-DPB1＊040101 allele is involved in susceptibility of developing this disease.HLA-DPB1＊0502-030101 and HLA-DQB1＊0202-020101 have a protective effect in chronic sinusitis with nasal polyps.
