益生注射液抗大鼠移植心脏慢性失功能加快模型的机制探讨

摘　　要：目的探讨中药益生注射液抗大鼠移植心脏慢性失功能加快模型的机制。方法采用雌性封闭群SD大鼠20只,随机数字表法分为2组,对照组的离体心脏用4℃生理盐水保存3h后行心脏移植,移植术后不给任何药物处理;实验组的离体心脏则用含益生注射液的4℃生理盐水保存3h后行心脏移植,术后每天腹腔注射益生注射液8mg/kg,连续7d。在移植术后第8天,取2组移植心脏进免疫组织化学染色,检测心肌组织中核因子-κB(NF-κB)、细胞黏附分子-1(ICAM-1)、Ⅱ类主要组织相容性复合物(MHC-Ⅱ)和转化生长因子-β1(TGF-β1)的表达情况。结果 2组心肌组织中均有NF-κB、ICAM-1、MHC-Ⅱ和TGF-β1的表达,但实验组4种分子的表达明显低于对照组(P<0.05)。结论在缺血-再灌注损伤诱导的移植心脏慢性失功能加快模型中,益生注射液对移植心脏有保护作用,其机制可能是减轻缺血-再灌注损伤所致的炎性级联反应。Objective To investigate the possible mechanism of the traditional chinese medicine Yisheng injection,which can reduce the progress of a novel accelerated rat cardiac graft failure model induced by ischemia/reperfusion injury.Methods 20 female closed colony SD rats were divided into two groups at random.In the control group,the isolated rat hearts were preserved in 0.9% sodium chlorine solution at 4℃ for 3 hours before transplantation,while in the experimental group,the hearts were preserved in 0.9% sodium chlorine solution containing Yisheng injection at 4 ℃ for 3 hours before transplantation.After transplantation,the experimental group recipients were treated with Yisheng injection 8 mg·kg-1·d-1 for 1 week.Grafts were excised on the 8 days after transplantation to detect the expression of NF-κB,ICAM-1,MHC-II and TGF-β1 using immunohistochemistry methods.Results We can detect the expressions of NF-κB,ICAM-1,MHC-II,and TGF-β1 by 8 day after transplantation in two groups.However,the expressions of NF-κB,ICAM-1,MHC-II,and TGF-β1 were significantly decreased in the experimental group.Conclusion It demonstrated that Yisheng injection can reduce the progress of a novel accelerated rat cardiac graft failure model induced by ischemia/reperfusion injury.Its possible molecular mechanism is that Yisheng injection could reduce the cytokine adhension molecules cascade.
