摘 要:目的:建立微波协助提取法提取黄芩中有效成分黄芩苷,比较微波协助提取法与药典法提取黄芩苷优势。方法:采用Dikma Diamonsil C18色谱柱(4.6 mm×150 mm,5μm),流动相为甲醇-水-磷酸(47∶53∶0.2),检测波长280nm,柱温30℃,流速1 mL.min-1。结果:微波提取时间15 min,提取温度120℃,提取溶剂30%乙醇。黄芩苷在0.059~0.59μg呈良好线性关系,r=0.999 9。平均回收率99.36%(n=6)。结论:微波提取法较药典法提取黄芩苷更简便快速、结果准确,为一种符合环保绿色理念的含量测定方法,可用于检测黄芩饮片中有效成分黄芩苷含量。Objective:To establish a microwave-assisted extraction(MAE)method for the extraction of the baicalin from Scutellaria baicalensis.Comparison of the determination of baicalin from S.baicalensis between MAE and Chinese pharmacopeia method.Method:Dikma Diamonsil C18 column(4.6 mm×150 mm,5 μm)was used.The mobile phase of methanol-water-phosphoric acid(47∶53∶0.2).The temperature of column was 30 ℃.The flow rate was 1.0 mL·min-1 and the detection wavelength was at 280 nm.Result:Microwave extractive time was 15 min,the temperature was 120 ℃,the extraction solvent was 60%ethanol.The calibration curve was in good linearity with in the range from 0.059 to 0.59 μg(r=0.999 9).The average recovery was 99.36%(n=6).Conclusion:The method is simple,rapid,as well as precise and reliable,and can be used for the determination of baicalin in S.baicalensis.
