中国北方汉族人群E-选择素第2外显子+G98T基因多态性与冠心病关系的研究

摘　　要：目的研究E-选择素(E-selectin)第2外显子+G98T基因多态性与中国北方汉族人群冠心病发病的关系。方法应用聚合酶链反应限制性片段长度多态性(PCR-RFLP)技术检测中国北方汉族无血缘关系的176名冠心病患者和152名健康对照者E-选择素基因第2外显子+G98T基因型;生化技术测定血脂水平;酶联免疫吸附试验(ELISA)双抗体夹心法检测血清E-选择素水平。结果 E-选择素+G98T基因型在冠心病组及对照组中的分布差异有显著性(χ2=4.628,P=0.031),GT基因型者患冠心病的风险是GG基因型的2.221倍(OR=2.221,95%CI=1.001～4.595);T等位基因高于G等位基因,差异具有显著性(χ2=4.351,P=0.037,OR=2.124,95%CI=1.047～4.310);中国北方汉族人群E-选择素基因+98位点T等位基因携带频率明显低于美国、德国人群(P<0.05)。结论 E-选择素基因第2外显子+G98T多态性与冠心病的发病具有相关性,T等位基因可能是北方汉族人冠心病发病的遗传易感基因,E-选择素基因+G98T引起的冠心病发病在一定程度上存在地域和种族差异。【Objective】To study the relationship between polymorphisms of E-selectin gene with coronary heart disease of Han population in northern China.【Methods】The ＋G98T polymorphism in the exon 2 of E-selectin gene was detected by polymorphisms chain reaction-restriction fragment length polymorphism（PCR-RFLP） analysis and DNA sequencing among 176 patients with coronary heart disease of Han people in northern China and 152 healthy controls.The effects of E-selectin genotypes on serum lipids variation were analyzed.The serum level of E-selectin was determined by enzyme-linked immunosorbent assay（ELISA）.【Results】There was significant difference in frequencies of allele and genotype in ＋G98T polymorphism between coronary heart disease and control groups,respectively（χ2=4.628,P =0.031）.The relative risk suffered from coronary heart disease of GT genotype was 2.221 times of the GG genotype（OR=2.221,95% CI=1.001～4.595）.Allele T was higher than allele G with significant difference（χ2=4.351,P =0.037,OR=2.124,95% CI=1.047～4.310）.【Conclusion】E-se1ectin ＋G98T polymorphism was associated with coronary heart disease,and allele T may be a genetic risk factor of coronary heart diseases among Han people,the E-selectin GT genotype carriers maybe different in different region and different race which results in coronary heart disease in some degree.
