摘 要:目的:研究N-乙酰半胱氨酸(NAC)对成年大鼠高氧肺损伤氧化应激机制的影响。方法:将48只健康成年雄性SD大鼠随机分为6组,其中5组置入动物实验氧舱内吸入100%浓度氧气,吸氧时间为24 h、48 h、72 h、96 h、96 h,作为24 h、48 h、72 h、96 h组和NAC干预组,还有1组吸入空气0 h作为0 h组,即对照组;计算氧合指数,测定肺系数;测定肺组织和血清中MDA含量,SOD及T-AOC活性;观察肺脏组织病理学变化。结果:随高氧暴露时间延长,大鼠氧合指数逐渐下降,肺系数逐渐增加。肺组织和血清中MDA含量逐渐增加,96 h达到峰值,SOD、T-AOC活性逐渐下降,96 h达到最低值。与96 h组比较,NAC干预组氧合改善,肺系数下降,MDA含量减少,SOD及T-AOC活性增加。结论:NAC通过减轻机体的氧化应激反应在一定程度上能够改善高氧所致成年大鼠急性肺损伤。Objective: To study the effects of N-acetylcysteine on oxidative stress in hyperoxia-induced lung injury of adult rats. Methods: Forty-eight healthy male SD adult rats were randomly divided into six groups: Five groups were put into oxygen chamber with oxygen concentration of 100% for 24 h, 48 h, 72 h, 96 h, 96 h, as 24 h, 48 h, 72 h, 96 h and NAC intervention group. Control group (0 h) breathed with room air. Arterial blood gases were measured to calculate oxygenation index. Lung coefficients were calculated. The MDA contents and SOD, T-AOC activities were assayed respectively in lung and serum. The changes of lung histopathology were observed. Results: The oxygenation index decreased gradually and lung coefficients increased gradually with contact time that rats were exposed to oxygen concentration of 100%. The contents of MDA in lung and serum reached maximal value at 96 h and SOD, T-AOC activities reached minimal value at 96 h. The oxygenation index was higher in N-acetylcysteine group than in 96 h group, lung coefficients and contents of MDA decreased, SOD, T-AOC activities increased in lung and serum. Conclusion: N-acetylcysteine can attenuate hyperoxia-induced lung injury of adult rats by inhibiting oxidative stress in some levels.
