摘  要:目的:探讨地高辛对小鼠吗啡行为敏化的影响。方法:测定小鼠的自主活动,观察地高辛对小鼠自主活动的影响。急性吗啡(10 mg.kg-1i,p)处理小鼠前给予地高辛,观察地高辛对急性吗啡引起小鼠高活动性的影响。慢性吗啡处理小鼠,建立吗啡行为敏化模型,观察地高辛对吗啡行为敏化的影响。结果:(1)地高辛(0.5 mg.kg-1-2.5 mg.kg-1i,g)呈剂量依赖地抑制小鼠本身的自发活动和急性吗啡诱导的小鼠高活动性;(2)地高辛剂量依赖性地阻断小鼠吗啡行为敏化的形成和表达。结论:地高辛对中枢神经系统具有抑制作用,并能够抑制吗啡引起的小鼠高活动性以及行为敏化的形成与表达,提示地高辛可能对吗啡的精神依赖性具有干预作用。Objective:To investigate the effects of digoxin on morphine induced behavioral sensitization.Methods:Locomotor activity was measured every 15 min for 2 hours after administration with digoxin in mice.The animals were rendered sensitization to morphine via treatment with morphine(10 mg ·kg-1,ip) for 5 days.After cessation of drugs for 7 days,all animals were challenged with morphine(10 mg ·kg-1,ip) on 12th day.Results:Digoxin dose-dependently inhibited both locomotor activity in mice and hyperlocomotion induced by acute morphine.Similar inhibition was also seen both in the development and the expression of morphine-induced behavioral sensitization.Conclusion:Digoxin inhibits both in the development and the expression of morphine-induced behavioral sensitization in mice.And the inhibitory effects of digoxin on dopamine release in brain may account for this.These findings indicate that digoxin may have the interferential potential to psychological dependence on morphine,and Na+,K+/ATPase might serve as a drug target in morphine addiction.
