摘 要:目的:建立高速逆流色谱分离纯化紫苏叶中迷迭香酸的方法。方法:采用高速逆流色谱分离纯化紫苏叶乙酸乙酯萃取部分中迷迭香酸,以石油醚-乙酸乙酯-甲醇-0.5%醋酸水溶液(2∶5∶2∶5)为溶剂体系,上相为固定相,下相为流动相,流速2.0 mL.min-1,主机转速800 r.min-1,温度25℃。结果:每0.20 g紫苏叶乙酸乙酯萃取物经1次纯化,纯度为98.6%的迷迭香酸的得率约13%。结论:高速逆流色谱可高效分离纯化紫苏叶中的迷迭香酸。Objective: To establish a method for separation and purification of rosmarinic acid from Perilla frutescens( L. ) Britt. vat. acuta ( Thunb. ) Kudo form. discolor( Makino ) Kudo ( abbr: PFAD) by high - speed counter - current chromatography( HSCCC ). Methods: The compound was separated and purified by HSCCC from the ethyl acetate part of the crude extract of PFAD, using petroleum ether - ethyl acetate - methanol - 0. 5 % acetic acid water -solution(2: 5: 2:5 )as the two -phase solvent system. The upper phase was chosen as the stationary phase, and the lower phase was chosen as the mobile phase. The mobile phase was operated at a flow rate of 2. 0 mL ·min^-1, while the apparatus rotated at 800 r ·min^-1 and the temperature was set at 25 ℃. Results: The yield of rosmarinic acid with 98.6% purity was 13% from each 0.20 g ethyl acetate part of the crude extract of PFAD by once separation. Conclusion: HSCCC is a efficient method for separating, and purificating rosmarinic acid from PFAD.
