摘  要:目的:研究三维培养环境下骨髓间充质干细胞的生长情况,探讨骨髓间充质干细胞复合支架构建组织工程软骨的可行性。方法:全骨髓培养法分离骨髓间充质干细胞(bone mesenchymal stem cells,BMSCs),免疫细胞化学测定其细胞表面抗原CD44、CD45,对BMSCs进行鉴定;冻干法制备壳聚糖(Chitosan,CS)支架,扫描电镜观测支架孔径;取BMSCs复合CS支架,通过三维培养观察BMSCs在CS支架上不同时段的生长情况。结果:三维培养环境下骨髓间充质干细胞生长良好,细胞在支架上分泌大量细胞外基质,并与支架黏附良好。结论:骨髓间充质干细胞可作为组织工程的种子细胞,构建组织工程软骨。Objective To study the growth of bone mesenchymal stem cells with the chitosan-collagen scaffolds.Methods BMSCs were isolated from bone marrow and cultured by general bone marrow methods.Both CD44 and CD45 on the BMSCs surface were detected with immune-cyto-chemistry to identify BMSCs.Chitosan scaffolds were produced by lyophilization and examined by SEM and observed the morphology of the BMSCs with chitosan scaffolds.Results In 3-D culture,cells in-growth in the pores,attaching and spreading on the inner and external surfaces of the scaffolds could be seen.A lot of cells had developed long cyto-plasmic extensions anchoring to the scaffolds.Conclusion BMSCs with CS scaffold grows well in 3-D culture,and the BMSCs can be used to chondral tissue engineering.
