摘  要:为了找出一种较为适宜的野生豹斑毒鹅膏菌的液体培养方式,对野生豹斑毒鹅膏菌进行了液体培养工艺及其产物毒性的初步研究,并以菌丝体生物量和小鼠毒性反应为指标,通过单因素试验和正交试验,对豹斑毒鹅膏菌的液体培养条件和培养基配方进行筛选及优化。结果表明:豹斑毒鹅膏菌的最佳液体培养基为20.0%马铃薯、2.0%葡萄糖、0.3%维生素液(VB10.05%,VPP0.25%,VB60.05%,VC0.05%,甘氨酸1%,蒸馏水定容)、0.3%硝酸钠、0.1%K2HPO4、0.05%氯化钾、0.05%MgSO4.7H2O、0.001%FeSO4,以蒸馏水定容,自然pH。最佳培养条件为:选取3d菌龄的菌丝体约1cm2,接入160mL装液量的锥形瓶中,26℃、120r/min暗培养8d,其菌丝体生物量为8.93g/L,毒性亦为最佳,可在较短的时间内导致小鼠出现神经精神型、肝损害型和胃肠炎型的综合性中毒反应,但主要以神经精神型为主,并在平均中毒时间为40.3h后导致小鼠死亡。The liquid culture condition and medium formula of A.pantherina and its product toxicity were studied by the single-factor and orthogonal tests.The results showed that the optimum medium for culture of A.pantherina was 20.0% potato,2.0% glucose,0.3% multivitamin(0.05% VB1,0.25% VPP,0.05% VB6,0.05% VC and 1% glycine),0.3%NaNO3,0.1% K2HPO4,0.05% KCl,0.05% MgSO4·7H2O and 0.001% FeSO4.The optimum culture condition with 8.93 g/L mycelium biomass and the best toxicity could be obtained under 1 cm2 mycelium with 3 d age in a conical flask with 160 mL,26℃,120 r/min and dark culture for 8 d.The fermentation A.pantherina liquor could result in the comprehensive toxic reaction of nerve-spirit,liver injury and gastroenteritis to mouse and the main toxic type was nerve-spirit.The mouse injected with 1 mL fermentation A.pantherina liquor would die after 40.3 h on average.
