MMP-2和TIMP-2mRNA在大鼠颈动脉粥样硬化中的动态表达

摘　　要：目的探讨基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶组织抑制剂-2(TIMP-2)在颈动脉粥样硬化(AS)发生、发展中的作用。方法将40只SD大鼠随机分正常对照组和钳-脂组(给予血管钳夹术和高脂饮食),钳-脂组又分为2、6、10周三个亚组,应用RT-PCR的方法,观察MMP-2、TIMP-2和MMP-2/TIMP-2 mRNA在各时间点颈总动脉中的动态变化。结果钳-脂各组大鼠颈总动脉的MMP-2和TIMP-2 mRNA逐渐增高,与正常对照组相比差异有统计学意义(P<0.05)。MMP-2/TIMP-2 mRNA:正常组<钳-脂2周组<钳-脂10周组,钳-脂6周组较其他三组低。结论在钳-脂法建立的大鼠颈AS病变中,随着AS的进展,MMP-2、TIMP-2 mRNA的表达逐渐增强,MMP-2/TIMP-2 mRNA除纤维斑块阶段外,其他阶段逐渐升高。Objective To explore the effect of matrix metalloproteinase-2（MMP-2） and tissue inhibitor of metalloproteinase-2（TIMP-2） in the occurrence and development of artery atherosclerosis（AS）.Methods Forty SD rats were divided into two groups randomly,the normal control group and the model group（high fat feeding and vascular clamping）.The model group was divided into three subgroups of 2 nd,6 th and 10th week.The dynamic variation of MMP-2,TIMP-2 and MMP-2/TIMP-2 mRNA were examined with the method of RT-PCR.Results The expression of MMP-2 and TIMP-2 mRNA in model group gradually increased,and was higher than normal group（P〈0.05）.MMP-2/TIMP-2 mRNA： normal group2 th week10 th week,the expression of 6 th week was lower than normal group.Conclusion The expression of MMP-2 and TIMP-2 mRNA gradually increase in model group,the expression of MMP-2/TIMP-2 mRNA in every phases is gradually increased except fibrous plaque.
