摘 要:目的:探讨20种常用生化试剂对测定:肌酐(CREA)、尿酸(UA)时产生交叉污染的项目及避免方法。方法:把20种常用生化试剂分别当标本进行CREA、UA两个项目的检测,筛查出对其检测存在污染的试剂,基于基质效应,进一步把一份混合血清分两份,一份加等量生理盐水,另一份加等量试剂当标本进行检测,确定交叉污染的情况。结果:谷丙转氨酶试剂1、谷草转氨酶试剂11、碱性磷酸酶试剂1、钙试剂、尿素氮试剂1、甘油三酯试剂1对肌酐,谷丙转氨酶试剂1、谷草转氨酶试剂、R-谷氨酰转移酶试剂1、尿素氮试剂2、肌酸激酶试剂1、磷试剂对尿酸存在不同程度的污染。结论:20中生化试剂中,有相当部分的试剂间对CREA、UA这两项目存在着一定的交叉污染情况,因此,在全自动生化分析仪上使用时,应注意项目分析顺序和试剂针特殊清洗程序的建立以避免交叉污染。Objective:To address the cross-contamination and solutions with biochemical analyzer measured outcomes of creatinine (CREA) and uric acid (UA) using 20 types of biochemical reagents. Methods: Of 20 common types, biochemical reagents that were frequently associated with contamination in measurement of CREA and UA were screened. Thereafter, a matrix effect-based test was conducted to evaluate the cross-contamination among samples of pooled serum which were split into duplicate aliquots, one added with an equal volume of normal saline and the other added with an equal volume of study reagent. Results : A range of cross-contamination was noted in measurement of creatinine using ALT reagent No. 1, ASP reagent No. 1, ALP reagent No. 1, calcium reagent, urea nitrogen reagent No. 1, and triglyeeride reagent no. 1, as was in measurement of uric acid using ALA reagent No. 1, AST reagent, R-glutamyltransferase reagent No. 1, urea nitrogen reagent No. 2, creatine kinase reagent No. 1, and phosphorus reagent. Conclusion : A range of cross-contamination among several of 20 types of common biochemical reagents may be present in measurements of CREA and UA. Therefore, establishing special algorithms for certain measurement and rinsing procedures of reagent probes may be important to reduce cross- contamination in measurements with automated biochemical analyzers.
