摘 要:目的:研究姜黄素对慢性应激模型大鼠空间学习记忆能力的影响及其可能机制。方法:随机分为正常组、应激模型组、姜黄素组(2.5、5和10 mg/kg,p.o.)和氟西汀(10 mg/kg,i.p.)组,每组8只。采用21 d慢性不可预知的刺激法建立大鼠应激模型,观察大鼠八臂迷宫和Y迷宫学习记忆成绩的改变,同时采用Western-blot等方法检测海马内脑源性神经营养因子(BDNF)和磷酸化cAMP反应元件结合蛋白(pCREB)的表达情况。结果:与正常对照组比较,应激模型组学习记忆能力明显下降(P<0.01),姜黄素组(5和10 mg/kg)能明显改善这一现象(P<0.05,P<0.01);应激模型组大鼠海马内BDNF和pCREB的含量较正常组显著降低(P<0.01),姜黄素组(5和10 mg/kg)与模型组比较含量有所提高(P<0.05,P<0.01),这一结果与阳性对照药氟西汀组相似。结论:姜黄素可以逆转慢性应激对大鼠学习记忆的损伤,这一作用可能与增强cAMP-CREB-BDNF信号转导通路,从而影响神经元存活及其可塑性机制有关。Objective: To investigate the offect of curcumin on learning memorial ability deficit in the chronically stressed rats and the its possible protective effects on learning-memory ability.Methods:Adult male Wistar rats were randomly divided into six groups: the control rats were housed in normal situations without any treatment;the chronically stressed rats were subjected to a variety of unpredictable stressor for 21 days;curcumin group(2.5,5,10 mg/kg,p.o)and fluoxetine group(10 mg/kg,i.p.).After the stress experiment,all rats were tested for eight-arm maze task and Y-maze spatial recognition memory.In addition,the possible mechanism of neurogenesis on stressed rats and the effects of curcumin were also analysed by western-blot and immunohistochemistry assay.Results: Compared with the control group,the stress induced mouse model decreased learning ability(P 0.001),curcumin(5,10 mg/kg)could improve the ability of learning memory(P 0.05,P 0.01).The expression of pCREB and BDNF in hippocampus was distinctively decrease in stressed rats(P 0.01),while curcumin(5,10 mg/kg) could reverse this condition.(P 0.05,P 0.01).Conclusion: Curcumin reverses the learning memory damage.These effects may be related to facilitaion of the cAMP-CREB-BDNF signal transduction cascades,which influences the neuron survival and adaptive plasticity induced by curcumin.
