摘  要:目的:检测人巨细胞病毒(HCMV)感染人胚肺成纤维细胞(HELF)后不同时间点HCMV DNA载量,以明确HCMV感染HELF的时效性。方法:将HCMV接种HELF细胞,收集感染后3,5,8,10,15,20,30,60,120 min时间点细胞,提取病毒DNA,以即刻早期1基因(IE-1)为靶基因进行荧光定量PCR检测。结果:在HELF受到感染的每个时间点细胞里都能检测到HCMV DNA,比较HCMV DNA拷贝数的对数发现,3,5,8,10,15,20 min组间比较及其与30,60,120 min比较,差异有统计学意义(P<0.01),而30,60,120 min之间比较,差异无统计学意义(P>0.05)。结论:HCMV感染HELF后3 min即可以进入细胞,随后病毒量随时间而递增,达到30 min后感染量基本达到平衡,随时间的递增感染量不再明显增加。Objective: To explore the human cytomegalovirus(HCMV) DNA load after HCMV infecting human embryo lung fibroblasts(HELF) in different time.In order to know the efficiency of the HCMV infecting HELF.Methods: We took HCMV to vaccinate HELF cells,and collected cells had been infected HCMV at the 3,5,8,10,15,20,30,60,120 min time point,and then extracted DNA of viruses.The early transcriptional regulation protein IE-1 was taken as target genes in fluorescence quantitative polymerase chain reaction(PCR) testing.Results: The HCMV DNA can be detected in the every time point cells of infected HELF.It was founded through the logarithm comparison of the HCMV DNA copy numbers of each time point of the infecting HELF that the comparison among 3,5,8,10,15,20 min groups and the comparison between them and 30,60,120 min groups,were statistics significantly difference(P 0.01),and the comparison among 30,60,120 min groups had no significant difference(P0.05).Conclusion: The HCMV can enter the cell after HCMV infecting HELF 3 min,and then the amount of HCMV infecting HELF increasing with time increasing,but the amount of HCMV infecting HELF was to be balanced up to 30 minutes,and then the increasing amount of HCMV infecting HELF was no longer obvious.
