川芎嗪对大鼠被动型Heymann肾炎肾小球细胞增生及细胞外基质分泌的影响

摘  要:目的:复制大鼠被动型Heymann肾炎(passive Heymann nephritis,PHN)动物模型,观察中药川芎嗪(Ligus-trazine)对PHN大鼠肾小球细胞增生和细胞外基质(extracellular matrix,ECM)分泌的影响。方法:给SD大鼠静脉一次性注射抗Heymann的抗血清建立PHN大鼠模型(设正常血清对照组),同时给大鼠腹腔注射川芎嗪注射液(20 mg/100 g体质量,2天1次直至第5周末),然后于注射抗血清后的1,3,5周末时,取大鼠的肾皮质制成切片,分别进行HE染色计数肾小球细胞总数;免疫组化染色检查肾小球内增殖细胞核抗原(proliferative cell nuclear antigen,PCNA)、α-平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)、Ⅳ型胶原纤维(collagenⅣ,Col-Ⅳ)和纤维黏连蛋白(fibronectin,FN)的表达;另行电镜观察肾小球系膜区超微结构的变化。结果:PHN+川芎嗪组肾小球细胞总数、PCNA和α-SMA的表达与PHN模型组相比,未显示出明显的降低(P>0.05),Col-Ⅳ和FN的表达则显著低于PHN模型组。电镜检查证实,PHN+川芎嗪组大鼠系膜区基质积聚明显减轻。结论:中药川芎嗪能有效抑制PHN大鼠肾小球内Col-Ⅳ、FN的产生,但对细胞增生的影响并不明显。Objective: Rat passive Heymann nephritis(PHN) was induced to explore the effects of traditional Chinese medicine Ligustrazine on the glomerular cell proliferation and extracellular matrix(ECM) secretion of the rats with PHN.Methods: SD rats were injected intravenously with anti-Heymann antigen antiserum to induce PHN,and meanwhile,the rats were also given Ligustrazine intraperitoneally(20 mg/100g BW,1 time / 2 days until the fifth weekend).The control group of rats were injected intravenously with normal rabbit serum.Then,the renal cortex of rats was taken on the 1,3 and 5th weekend after the antiserum injection,and the sections were used to perform HE staining to count the number of total glomerular cells.The glomerular expression of proliferating cell nuclear antigen(PCNA),α-smooth muscle actin(α-SMA),collagen Ⅳ(Col-Ⅳ) and fibronectin(FN) were also determined by immunohistochemical staining.In addition,the mesangial ultrastructural changes were observed under electron microscopy.Results: PHN rats treated with ligustrazine(ie,PHN + ligustrazine group) exhibited less expression of Col-Ⅳ and FN compared with PHN rats,although there was no significant difference in the number of total glomerular cells,and the expression of PCNA and α-SMA between two groups.Moreover,rat mesangial ECM accumulation was reduced in PHN + ligustrazine group compared with PHN model rats by using electron microscopy.Conclusion: Ligustrazine can inhibit the glomerular production of Col-Ⅳ and FN,but not cell proliferation in PHN rats.
