摘  要:[目的]观察痛泻要方及拆方对腹泻型肠易激综合征(D-IBS)模型大鼠结肠组织水通道蛋白3(aquaporin3,AQP3)表达的影响,探讨痛泻要方的作用机制。[方法]采用慢性应激与束缚相结合方法建立D-IBS大鼠模型;免疫组化法检测AQP3含量;RT-PCR法检测AQP3mRNA的表达。[结果]与对照组比较,模型组和痛泻要方组结肠组织中AQP3表达显著下调(P<0.01);与模型组比较,痛泻要方与方中去陈皮组的结肠组织中AQP3表达均上调(P<0.01);而与方中去白术或去白芍组差异无统计学意义。[结论]痛泻要方中"补脾止泻"药主要是白术和白芍,并且其补脾止泻的作用机制之一可能与上调结肠组织AQP3表达有关。[Objective]To observe effect of Tongxieyaofang and its combining formula on the expression of colon tissue aquaporin 3(AQP3)in diarrhea irritable bowel syndrome(D-IBS)rat models. [Methods]D-IBS model was set up by using the combining method of chronic stress and constraint.AQP3 content was detected with immunohistochemistry method and its mRNA expression was measured by RT-PCR. [Results]Compared with the control group,AQP3 expression in model group and Tongxieyaofang group decreased significantly(P0.01).Compared with the model group,AQP3 expression in Tongxieyaofang group and its group without Pericarpium Citri Reticulatae increased significantly(P0.01).Groups without Rhizoma Atractylodis Macrocephalae or Radix Paeoniae Alba had no significant difference. [Conclusion]Chinese materia medica of reinforcing spleen to relieve diarrhea in Tongxieyaofang are mainly Rhizoma Atractylodis Macrocephalae and Radix Paeoniae Alba.One of its anti-diarrhea mechanism is possibly related to the increase of colon tissue AQP3 expression.
