摘 要:目的探讨营养性肥胖对青春期大鼠睾丸生殖细胞中Fas/Fas-L凋亡途径的影响。方法24只3周龄健康青春期Wistar雄性大鼠,按随机化原则分为对照组和高脂组两组,每组12只,对照组给予普通饲料喂养,高脂组用高脂、高热量饲料喂养,10周末分别测定大鼠体重,随后处死大鼠采集血标本摘取睾丸。全自动生化分析仪(ACA)检测外周血总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL—C)。光镜观察睾丸组织病理学改变,原位缺口末端标记法(TUNEL)检测睾丸组织凋亡细胞,免疫组化法检测Fas、Fas-L和Caspase-3蛋白的分布和表达。结果高脂组TC、TG、LDL-C和HDL-C较对照组明显升高,差异有统计学意义(P〈0.05);高脂组生殖细胞凋亡指数明显高于对照组(P〈0.01),凋亡细胞以精原细胞和精母细胞为主;高脂组Fas、Fas-L和Caspase-3蛋白表达明显高于对照组(P〈0.01)。结论营养性肥胖可诱导大鼠睾丸生殖细胞凋亡增多,其机制可能与Fas、Fas-L和Caspase-3蛋白的表达升高有关。Objective To investigate the effects of dietary-induced obesity on the Fas/Fas-L apoptosis pathway oftesticular germ cells of pubertal rats. Methods Twenty-four health male rats were randomly divided into the control group and the high-fat group. The rats in the control group (n=12) were fed on normal diet; and the rats in the high-fat group (n=1 2) were given high-fat and high-calorie diet. All rats were killed at the tenth weeks, and their testis were obtained. Levels of peripheral blood total cholesterol (TC), Triglyceride (TG), High-density lipoprotein cholesterol (HDL-C) and Low-density lipoprotein cholesterol (LDL-C) were detected by automatic biochemical analyzer (ACA). The pathological changes of testis were observed by light microscope. The apoptosis of testis cells was detected by TUNEL staining. The expressions ofFas, Fas-L and Caspase-3 protein in testis were detected by immunohistochemistry. Results Levels of serum TC, TG, LDL-C and HDL-C in the high-fat group were obviously higher than those in the control group(P〈0.05), The apoptotic index of germ cells in the high-fat group was significantly higher than that in the control group(P〈0.01). The apoptotic germ cells were mainly spermatogonia and spermatocytes. The expressions of Fas, Fas-L and Caspase-3 protein in the high-fat group were obviously higher than those in the control group(P〈0.0 1). Conclusion Alimentary obesity might induce apoptosis of germ cells of testis, and its mechanism might be associated with upregulation ofFas, Fas-L and Caspase-3 protein
