摘　　要：目的检测三氯乙烯(TCE)染毒小鼠体内Th17细胞数量及其分泌的白介素17(IL-17)含量和肝功能的变化,探讨Th17细胞与TCE引起的肝功能损害的关系。方法雌性BALB/c小鼠随机分为TCE(2.5、5 mg/ml)组、空白对照组和溶剂对照组。染毒2、4、8、12周,采集外周血用全自动生化仪测定肝功能,处死动物取脾脏和肝脏,流式细胞仪检测脾脏Th17细胞数量,ELISA法检测肝组织匀浆中IL-17含量。结果与对照组相比,TCE组小鼠天冬氨酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT)水平明显升高,2周和4周时差异有显著性(P<0.05);肝组织匀浆中IL-17含量与对照组比较,TCE组也明显升高(P<0.05,P<0.01),4周时最高;与对照组比较,TCE组小鼠脾脏Th17细胞数量明显升高,2、4、8、12周时差异均有显著性(P<0.05,P<0.01);相关分析显示肝功能与肝组织匀浆中IL-17含量和Th17细胞数量均呈正相关。结论 TCE引起的肝功能损害可能与Th17细胞有关。Objective To investigate the relationships of Th17 cells with liver functions damage in trichloroethylene（TCE） exposed mice.Methods Female BALB/c mice were intaked TCE（2.5,5 mg/ml） by drinking water;water and DMSO were served as blank and vehicle controls.At 2,4,8,12 weeks,blood were collected and assayed liver functions;liver and spleen biopsies were taken;Th17 cells number in spleen were measured by flow cytometry;IL-17 contents in liver homogenates were evaluated by ELISA.Results Compared with the control group,TCE treated groups AST and ALT increased,2 weeks and 4 weeks were significantly;IL-17 contents in liver homogenate of TCE treated group also significantly increased,at 4 weeks maximum;Th17 cells number in TCE treated mice spleen were significantly higher than these of control groups,there were significant differences at 2,4,8,12 weeks;correlation analysis showed that there were significantly positive correlation of liver functions with IL-17 contents in liver tissue and Th17 cells number in spleen.Conclusion These results suggest that the liver functions damage caused by TCE exposure may be related to Th17 cells.
