大鼠饮食相关性高草酸尿症和高钙尿症的实验研究

摘  要:目的探讨饮食钙和饮食草酸两因素对大鼠24小时尿草酸总排泄量和24小时尿钙总排泄量的影响。方法将24只大鼠随机分为低剂量钙组(A组)、中等剂量钙组(B组)及高剂量钙组(C组),连续喂养4 d,每组在4 d内分别依次给予剂量递减的饮食草酸。测量大鼠每天的24小时尿量、尿草酸浓度、尿钙浓度、尿肌酐浓度并计算24小时尿草酸总排泄量和24小时尿钙总排泄量。其中,尿草酸浓度用HPLC法检测,尿钙浓度和尿肌酐浓度用自动生化分析仪检测。用重复测量方差分析和多元方差分析对结果进行统计。结果对24小时尿草酸总排泄量在不同时间点比较,第3天和第1天相比下降(P=0.010),第4天和第1天相比下降(P=0.000),第4天和第2天相比下降(P=0.034),第4天和第3天相比下降(P=0.032),其余差异未见有统计学意义。对24小时尿钙总排泄量在不同时间点比较,第2天和第1天相比差异有统计学意义(P=0.037),其余差异未见有统计学意义(P>0.05)。结论 24小时尿草酸总排泄量随饮食钙摄入量提高而降低,随饮食草酸摄入量降低而降低。适当范围内补钙对24小时尿钙排泄量影响不大,而过度补钙可能会引起高钙尿症。Objective To explore the effect of the intake of calcium diet and oxalate diet on the output of urinary oxalate and urinary calcium of rat in 24 hours.Methods Twenty four SD male rats were randomly divided into low calcium diet group,medium calcium diet group and high calcium diet group,each group was sequentially fed on different oxalate diet in 4 days.The urinary volume in 24 hours was recorded,the consistency of urinary oxalate was detected by HPLC(high efficiency liquid chromatography),the consistency of urinary calcium and urine creatinine were determined by automatic biochemical analyzer.The consistency should be corrected to the output of urinary oxalate and urinary calcium of rats in 24 hours,the results were evaluated by repeated measurement of variance analysis and multivariate analysis of variance.Results d3 compared with d1,the output of urinary oxalate of rats in 24 hours decreased significantly(P=0.010),d4 compared with d1,the output of urinary oxalate decreased significantly(P=0.000),d4 compared with d2,the output of urinary oxalate decreased(P=0.034),d4 compared with d3,the output of urinary oxalate decreased(P=0.032).d2 compared with d1,the output of urinary calcium of rats in 24 hours decreased(P=0.037),and there were no significant difference in other days(P0.05).Conclusion The output of urinary oxalate of rats in 24 hours gradually decreased along with the increasing of the calcium diet.The output of urinary oxalate of rats in 24 hours gradually decreased along with the decreasing of the oxalate diet.Proper intake of calcium diet had no significant effect on urinary calcium excretion,but excessive intake of calcium diet could possibly lead to hypercalciuria.
