摘 要:利用TCA-丙酮沉淀-裂解液溶解法提取朗德鹅肝脏蛋白质,然后采用常规双向电泳技术对提取的蛋白质进行分离,利用PDQuest软件分析电泳图谱,统计蛋白质点及比较凝胶重复性。试验得到(712±15)个蛋白质点,蛋白主要集中在pI4.0-7.0和43.O~97.4ku,重复胶的匹配点数为694个,蛋白质点匹配率为96%,蛋白量的相关系数为0.875。本研究建立了朗德鹅肝脏蛋白双向电泳技术,双向电泳图谱中蛋白位点的分辨率和重复性较高,为进一步研究其蛋白质组学奠定了基础。Proteins were extracted by TCA/Acetone precipitation from liver tissue of Landes goose and separated by the two-dimensional electrophoresis(2-DE). Then 2-DE images were analyzed by PDQuest. The result indicated that TCA/Acetone precipitation was suitable for extraction and purification of protein from goose liver. 2-DE image analysis revealed (712±15)protein spots,and pI value was mainly ranged from 4.0 to 7.0 and molecular weight was ranged from 43.0 to 97.4 ku. The matching spots were 694 in two repeated eleetrophoregrams,with matching rate of 96%. The method of 2-DE of Landes goose's liver protein had been successfully established with high quality and resolution, which was valuable for proteomics research of Landes goose's liver.
