摘  要:目的研究氯化两面针碱对肝癌HepG2裸小鼠移植瘤的的抗肿瘤作用及机制。方法观察氯化两面针碱对肿瘤重量、体积的影响,电镜观察肿瘤细胞超微结构改变,TUNEL法分析其对细胞凋亡的影响。结果氯化两面针碱2.5,5,10mg.kg-1剂量对肝癌HepG2的抑制率分别为12.06%,35.63%和60.91%,电镜下可见NC给药组肿瘤细胞核染色质浓缩并边缘化,核碎裂,胞浆空泡化;TUNEL染色凋亡细胞明显增多,凋亡指数为(27.5±3.6)%。结论氯化两面针碱可以抑制肝癌HepG2的生长,其机制与促进肿瘤细胞凋亡有关。Objective To investigate the anti-tumor effect of nitidine chloride on human HepG2 hepatocellular transplanted tumor in nude mice and its mechanism.Methods The anti-tumor effect of nitidine chloride was evaluated by tumor weight and volume.The ultrastructural changes of HepG2 cells were examined with transmission electron microscope(TEM),and the apoptosis of tumor organization was detected by TUNEL assay.Results Nitidine chloride significantly inhibited the growth of tumor.The inhibitory rates at the dose of 2.5,5,10 mg·kg-1 were 12.06%,35.63% and 60.91%,respectively.Condensation of chromatin at margins of nuclei,disintegration of nucleolus,vacuoles in cytoplasm were observed in nitidine chloride group by TEM.The apoptosis index(AI) was(27.5±3.6)% detected by TUNEL assay.Conclusion Nitidine Chloride can inhibit hepatic carcinoma growth in nude mice,and the anti-cancer mechanism is probably associated with the induction of cancer cell apoptosis.
