摘  要:目的建立高效液相色谱(HPLC)法同时测定制何首乌中二苯乙烯苷、黄豆苷元、大黄素、大黄素甲醚的方法。方法采用高效液相色谱法;色谱柱为Diamonsil C18(5μm,250 mm×4.6 mm);柱温35℃;流速1.2 ml/min;检测波长280nm;流动相为甲醇∶0.5%醋酸溶液(42∶58)。结果被测定峰与其他组分峰完全分离。二苯乙烯苷、黄豆苷元、大黄素甲醚和大黄素的线性范围分别为0.908 4~9.084μg(R2=0.999 8),0.1236~1.236μg(R2=0.999 7),0.6048~6.048μg(R2=0.999 9),0.3648~3.648μg(R2=0.999 2);平均回收率(n=6)分别为100.2%,(RSD=1.9%),98.53%(RSD=2.0%),100.7%(RSD=2.8%)和99.95%(RSD=2.0%)。结论该方法准确、简便、重复性好,可作为制何首乌中二苯乙烯苷、黄豆苷元、大黄素、大黄素甲醚的含量测定方法,并易于推广。Objective To establish HPLC method for the simultaneous derermine of stilbene glycoside,daidzein,emodin and physcion in Radix Polygonum multiflorum.Methods The high-performance liquid chromatography was adopted.The chromatographic column was Diamonsil C18(5μm,250mm×4.6mm);Column temperature was 35℃,velocity was 1.2ml/min,the detection wavelength 280 nm,and the mobile phase was Methanol: 0.5% acetum(42∶58).Results The measured peak was completely separated from the other components in the peak.The linear ranges of stilbene glycoside,daidzein,physcion and emodin were 0.908 4~9.084 μg(R2=0.999 8),0.123 6~1.236μg(R2=0.999 7),0.604 8~6.048 μg(R2=0.999 9),0.364 8~3.648μg(R2=0.999 2),respectively.The average recoveries(n=6) were 100.2%,(RSD=1.9%),98.53%(RSD=2.0%),100.7%(RSD=2.8%)和99.95% respectinely(RSD=2.0%).Conclusion The method is accurate,simple,reproducible and can be used as assay of stilbene glycoside,daidzein,emodin and physcion in Radix Polygoni Multiflori Preparata.
