温下肠腑方对Lewis肺癌PCNA、p16、CyclinD1蛋白表达的影响

摘　　要：目的观察温下肠腑方对Lewis肺癌移植瘤的抑制作用及对PCNA、p16、cyclinD1表达的影响。方法:C57BL/6小鼠皮下接种Lewis肺癌,随机分为4组,即模型组,化疗组,温下方组和综合治疗组,化疗组和综合治疗组DDP 0.1 mg腹腔注射3 d,温下方组和综合治疗组中药灌胃10 d。计算抑瘤率,免疫荧光组织化学方法,激光共聚焦显微镜观察PCNA、p16、cyclinD1的表达。结果温下方组抑瘤率为34.5%,平均瘤重低于模型组(P<0.05);温下方组、综合治疗组和化疗组PCNA、cyclinD1表达明显低于模型组(P<0.05),其余各组无显著差异。结论温阳除滞法为主的温下肠腑方有一定的抑制肺癌生长的作用,且与顺铂有协同作用,温下肠腑方可能通过对PCNA、cyclinD1的抑制,从而抑制肿瘤生长。Objective To observe the inhibitory effect of Wenxiachangfu Decoction on the growth of C57BL/6 mice with trasplanted leweis tumor and on the expression of PCNA,p16、 and CyclinD1.Methods C57BL/6 mice with transplanted Lewis tumor were randomly divided into four groups：the model group（group A）,and chemotherapy treated group（group B）,Wenxiachangfu Decoction treated group（group C）,chemotherapy plus Wenxiachangfu Decoction treated group（group D）：Group B and D were treated with DDP 0.1mg injection in abdomen for 3 days,and group C and D were treated with Wenxiachangfu Decoction for 10 days.The inhibition rate was calulated and immunofluorescence histochemistry with the laser scanning confocal microscope was used to detect the expression of PCNA,p16,and cyclinD1.Results The inhibition rate of tumor in group C was 34.5%,the average tumor weight had significant difference as compare to group A.The laser scanning confocal microscope study showed the expression level of PCNA and cyclinD1 in group B,C and D was significantly lower than that of group A（P0.05,P0.05）.Conclusion Wenxiachangfufang Decoction has good effect in inhibiting the growth of lung cancer by regulating PCNA and cyclinD1 expression.
