2型糖尿病大血管病变患者ApoE基因多态性与中医证候的相关性研究

摘　　要：目的研究2型糖尿病(T2DM)大血管病变患者载脂蛋白E(ApoE)基因多态性与中医证候的相关性。方法 60例T2DM大血管病变患者按中医证候分为阴虚燥热、气阴两虚、阴阳两虚3组各20例,另设正常对照组20例,用聚合酶链反应扩增技术和限制性片段长度多态性技术(PCR-RFLP)对ApoE进行分析,并对患者的年龄、性别、体质量指数、病程进行分析。结果气阴两虚组患者ε4/4基因型所占比例及ε4等位基因频率、ε3/4基因型和ε3等位基因频率均有所升高(P<0.05)。T2DM大血管病变患者中E4组患者的病例数最多,E4组的病程与E2组比较,有显著的延长(P<0.05)。结论 T2DM大血管并发症患者气阴两虚型与ApoE基因ε4、ε3有内在关系,ApoE基因的多态性在一定程度上可以反应T2DM大血管病变患者的中医证候分型。Objective To research the correlation ship between symptom and sign of traditional Chinese medicine（TCM） and Apolipoprotein E genetype in type 2 diabetes mellitus with macrovascular complications.Methods 60 patients with type 2 diabetes mellitus macrovascular complications were divided into 3 groups as dryness-heat due to ＂Yin＂ deficiency group,deficiency both ＂Qi＂ and ＂Yin＂ group and deficiency both ＂Yin＂ and ＂Yang＂ group according to symptom and sign of TCM,20 cases in each group.ApoE genetype was estimated by polymerase chain reaction（PCR） and restriction fragment length polymorphisms（RFLP）,and patient＇s age,sex,body mass index,duration were analyzed.Results The frequencies of ε4/4、ε3/4 genotypes and ε4、ε3 in deficiency both ＂Qi＂ and ＂Yin＂ group were higher than those in dryness-heat due to ＂Yin＂ deficiency group,deficiency both ＂Yin＂ and ＂Yang＂ group and control group.The cases in E4 group was the largest number.The duration in E4 group was longer than that of E2 group.Conclusion There is inherent relationship between the symptom and sign of traditional chinese Medicine and apolipoprotein E genetype in Type 2 Diabetes mellitus with macrovascular complications.ApoE gene polymorphismε3、ε4 has a tight connection in Deficiency both ＂Qi＂ and ＂Yin＂ group in Type 2 diabetes mellitus with macrovascular complications.
