西南山茶种子、果皮、花抗IgE介导Ⅰ型过敏的研究

摘　　要：目的以致敏大鼠血清免疫球蛋白E(IgE)、白三烯(LT)和组胺(His),全血和肺泡灌洗液(BALF)中的嗜酸性粒细胞(EOS)和肺组织炎症面积为活性指标,跟踪研究西南山茶种子、果皮、花抗IgE介导Ⅰ型过敏有效部位和有效成分。方法西南山茶花用乙醇提取,提取物用醋酸乙酯萃取,醋酸乙酯萃取物进行化学成分分离。花用50%丙酮提取,种子和果皮分别用乙醇提取。用卵白蛋白(OVA)和氢氧化铝Al(OH)3的混合液致敏和诱发,制成Ⅰ型过敏反应动物模型。将实验动物随机分组即正常组,模型组,孟鲁司特钠组(Menglusitena)组,各样品的高、低剂量组。采用ELISA法测定血清IgE、LT和His含量;采用细胞计数法,测定大鼠全血和BALF中EOS数量;通过病理学检查,检测肺组织炎症面积。结果得到花的95%乙醇提取物(1),花的50%丙酮提取物(2),花的醇提物水层(3),化学分离得到5个黄酮类化合物,其中山萘酚(4)含量较高,种子的醇提物(5),果皮醇提物(6)。1,2,3,5,6号均可降低血清IgE、LT和His含量,强弱顺序5、6、1>2、3>4;1、2、5可明显减少全血及BALF中EOS数量,强弱顺序为1,2,5>3,6>4;1,3,5,6均可减少肺组织炎症面积,强弱顺序5>6>1>3。结论 1,2,3,5,6号灌胃给药对大鼠过敏均有明显的拮抗作用。综合各项指标,活性强弱顺序为5、6、1>2、3>4。Objective To track and study the effective part and constituent from flower,seed and the skin of fruit in Camellia pitardii Coh.St.Sealy in the anti-allergy of IgE involved in type I according to the activities with the serum sensitized serum IgE,LT,His,EOS in whole blood and BALF,inflammation in pulmonary tissue of sensitized rats. Methods The flower was extracted by EtOH and 50% acetone in order and EtOAc extract of EtOH extract were isolated by silica gel column.The seed and skin of fruit were extracted by EtOH.Rats were sensitized with the mixture of OVA and Al（OH）3 to establish the experimental animal model of type I allergy.The animals were divided into five groups randomly,namely normal group,model group,Menglusitena group,sample（low and high dose） groups.The IgE,LT,and His contents in serum of sensitized rats were measured with ELISA method.The inflammatory area in pulmonary tissue was detected by the pathological examination. Results EtOH extract（1）,50% acetone extract（2）,H2O layer（3）,Kaempferol（4）,EtOH extract of seed（5）,EtOH extract of skin of fruit（6） were obtained.1,2,3,5,6 could decrease the contents of IgE,LT and His in serum,the active sequence was 5,6,1 2,3 4;1,2,5 could reduce the quantities of EOS in whole blood and BALF,the active sequence was 1,2,5 3,6 4;1,3,5,6 could reduce the inflammatory area in pulmonary tissue,the active sequence was 5 6 1 3. Conclusion 1,2,3,5,6 have obvious anti-alergy effect by gastric administration,and the active sequence is 5,6,1 2,3 4.
