摘  要:目的探讨何首乌饮(HSWY)预防用药对雄性衰老大鼠生殖功能的保护作用。方法选用8周龄雄性SD大鼠60只,体质量180~220 g,随机分为正常对照组、模型组、何首乌饮预防中剂量(Pz)、低剂量(Pd)组,每组15只。除正常组外其余各组大鼠均采用D-半乳糖连续腹腔注射建立亚急性衰老大鼠模型,预防组在腹腔注射D-半乳糖的同时灌胃何首乌饮,用药60 d。检测各组血清胰岛素样生长因子-1(IGF-1)、黄体生成素(LH)、促卵泡激素(FSH)、睾酮(T)及下丘脑促性腺激素释放激素(GnRH)的含量;采用原位杂交法观察睾丸组织IGF-1 mRNA的表达;免疫组织化学法观察睾丸组织促卵泡激素受体(FSHR)、黄体生成素受体(LHR)、表皮细胞生长因子(EGF)及受体(EGFR)的表达;观察各组大鼠精子的数量、活动度和存活率的变化。结果模型组大鼠血清中LH、FSH及下丘脑中GnRH含量明显升高,IGF-1、睾酮含量明显下降,与正常组比较差异有显著性(P<0.05);何首乌饮中、低剂量组均能不同程度降低大鼠血清LH、FSH及下丘脑中GnRH含量,提高IGF-1、睾酮浓度,与模型组比较,差异显著(P<0.05)。模型组大鼠睾丸组织IGF-1mRNA、FSHR、LHR、EGF、EGFR的表达以及精子数量、活动度和存活率明显低于正常组(P<0.01);何首乌饮预防用药组能逆转上述现象,显著高于模型组(P<0.05)。结论何首乌饮通过改善激素的分泌,提高睾丸组织促性腺激素受体、生长因子及受体的表达,从而预防睾丸组织衰老,对雄性衰老大鼠生殖功能有较为明显的保护作用。Objective To study the protection of Heshouwuyin(HSWY) pretreatment on the reproductive functions of male aging rats. Methods Sixty 8-week-old male SD rats(weight:180~220 g) were randomly divided into 4 groups,15 rats in each: normal control group,aging model group,HSWY middle dosage group and HSWY low dosage group.Except the normal control group,the rats of the rest groups were all made by injecting D-gal into abdominal cavity continually.At the same time,the preventive graps were treated with intragastric administration of HSWY for 60 days.To detect the content of IGF-1,LH,FSH,T in the blood and GnRH in the hypothalamus.Hybridization in situ was used to observe the expression of IGF-1 mRNA in the testes of rats and immunohistochemical method was used to observe the expression of FSHR,LHR,EGF and EGFR in the testes of rats.The changes of the number,activity and the survival rate of the spermatozoa of rats were observed. Results LH,FSH and GnRH contents in model group were higher than normal group(P0.05),and those in HSWY preventive group were lower than model group(P0.05);IGF-1 and Ts contents in model group were lower than normal group(P0.05),and those in HSWY preventive group were higher than model group(P0.05).IGF-1mRNA,FSHR,LHR,EGF and EGFR contents in testicular tissue,spermatozoa number,spermatozoa activity and survival rate in model group were lower than normal group(P0.01),and those in HSWY preventive group were higher than model group(P0.05). Conclusion HSWY can increase the expression of GnRH-R and GF in testicular tissue and prevent the testes aging through improving the secret of sex hormone,which has obvious protection on the reproductive functions of male aging rats.
