摘  要:目的通过高效液相-电化学检测器(HPLC-ECD)检测大鼠纹状体细胞外液中去甲肾上腺素(NE)神经递质的变化,探讨松果菊苷(ECH)对脑神经保护作用的可能机制。方法 40只SD大鼠随机分为对照组、模型组、ECH高、低剂量组和川芎嗪组(CXQ)。各组大鼠给予相应的药物或生理盐水腹腔注射,1次/d,连续7 d,末次给药1 h后,制作大鼠局灶性脑缺血模型(MCAO),造模后立刻进行微透析,HPLC-ECD测定各组纹状体细胞外液中去甲肾上腺素的含量。结果与对照组相比,模型组动物纹状体细胞外液中NE含量先升高,后下降;而ECH高、低剂量组和川芎嗪(CXQ)组可以抑制NE减少,ECH高剂量组、CXQ组与模型组相比具有统计学意义(P<0.05或P<0.01)。结论实验建立测定脑内NE的方法简便,准确。ECH对增加脑缺血大鼠纹状体细胞外液中的NE含量释放具有较好的作用。Objective To examine the effects of echinacoside on striatal extracellular levels of norepinephrine in cerebral ischemia rats by HPLC-ECD and its possible mechanisms.Methods 48 rats were randomly divided into control,model,high,low and middle doses of echinacoside groups.Every group was injected into the right striatum of rats except the control group which was given equal volume of 0.9% saline,drugs or vehicle were administered one time a day for seven consecutive days.Focal ischemia was generated by permanent middle cerebral artery occlusion(MCAO).All rats in conscious and freely moving state using cerebral microdialysis technique.The change of extracellular NE levels in striatum of MCAO rats was observed dynamically.Results The results showed that the level of NE increased,then decreased rapidly,intraperitoneal injection of echinacoside could inhibit the decrease of extracellular NE level,and there were significant differences between ECHB,CXQ groups and model group(P 0.05,P 0.01 vs model group).Conclusion Our findings demonstrated that echinacoside can increase the striatal extracellular level of NE in the awake freely moving rats of cerebral ischemia injury.
