摘 要:目的探讨川芎、白芍及川芎-白芍配伍治疗原发性痛经的作用机理。方法利用原发性痛经模型小鼠,观察药物对小鼠40 min内扭体次数、扭体发生率的影响;测定小鼠子宫组织中一氧化氮(nitric oxide,NO)和钙离子的含量。结果与模型组比较,各给药组均可降低小鼠扭体次数(P<0.05或P<0.05),作用强度依次为白芍组>川芎组>川芎-白芍组;升高子宫组织中NO水平(P<0.05或P<0.01),作用强度依次为川芎组>白芍组>川芎-白芍组;显著降低子宫组织中钙离子水平(P<0.01),作用强度依次为白芍组>川芎组>川芎-白芍组。结论川芎、白芍及川芎-白芍药对对缩宫素所致痛经小鼠扭体有显著抑制作用,可升高子宫组织中NO水平,降低钙离子水平。这些作用可能与其治疗原发性痛经的部分机制有关。Objective To assess the effect of Rhizoma Chuanxiong and Radix Paeoniae Alba and the drug-pairs on primary dysmenorrhea and its mechanism.Methods Oxytocin and estrogen were employed to construct animal model of primary dysmenorrhea.The twisting times of mice in 40 min was measured,and the levels of nitric oxide and calcium ion in uterine tissue of mice were determined.Results Administration of Rhizoma Chuanxiong and Radix Paeoniae Alba and the drug-pairs obviously reduced the twisting times induced by oxytocin compared with negative control(P0.05).Rhizoma Chuanxiong and Radix Paeoniae Alba and the drug-pairs could increase nitric oxide concentration(P0.05) and obviously reduce calcium ion concentration in uterine tissue of mice(P0.01).Conclusion Rhizoma Chuanxiong and Radix Paeoniae Alba and the drug-pairs have certain pharmacological effects on primary dysmenorrhea and the mechanisms may be related with the increase of nitric oxide level and reduce of calcium ion concentration in uterine tissue of mice.
