摘 要:目的筛选滨蒿抗流感病毒的活性部位。方法体外实验测定滨蒿不同活性部位对MDCK细胞的半数有毒浓度(TC50)。观察样品对甲、乙型流感病毒致细胞病变(CPE)的抑制作用,计算半数抑制浓度(IC50)和选择指数(SI)。体内试验用流感病毒鼠肺适应株(PR8-M5)感染小鼠模型,以死亡率、生命延长率为指标,比较滨蒿不同活性部位体内抗流感病毒的作用。结果滨蒿H4部位在体外对甲、乙型流感病毒引起的细胞病变均有明显抑制作用,其IC50分别平均为35.3,84.0μg/ml,SI为9.9,4.1;其在体内给药剂量为0.4 g/kg时可明显延长感染小鼠存活天数,降低死亡率。结论滨蒿H4部位在体内外均具有明显的抗流感病毒作用。Objective To screen the anti-viral active site from Artemisia scoparia Waldst.et Kit.Methods The cell culture technique in vitro was used in MDCK cells to get TC50 of different active sites from Artemisia scoparia Waldst.et Kit.50% in hibitor concentration(IC50) and the select index(SI) were mesasured by cytopathic effect(CPE).The mice were infected with influenza virus PR8-M5 from nose.The death rate and the mean survival days of infected mice were observed to evaluate the therapeutic efficacy of different active sites from Artemisia scoparia Waldst.et kit.Results The cytopathic effect of the active H4 included by influenza virus was inhibited remarkably,with IC50 of 35.3,84.0 μg/ml and the SI of 9.9,4.1.At the dose of 0.4 g/kg body weight the survival rate was increased remarkably.Conclusion The active H4 of Artemisia scoparia Waldst.et Kit exhibits both anti-influenza activity in vitro and in vivo.
