大蓟醇提物对两肾一夹高血压大鼠Intermedin变化的影响

摘　　要：目的探讨大蓟提取物对两肾一夹(2-kidney1-clip,2K1C)高血压大鼠Intermedin(IMD)变化的影响。方法采用2K1C复制肾性高血压大鼠模型;检测心功能、左心重与体重比值(LVM/BW)及右肾重与左肾重比值(RK/LK);免疫组化法检测心脏、肾脏和腹主动脉IMD的水平;放射免疫学方法检测血浆中IMD和血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)的含量;采用硝酸还原酶法测定血清中一氧化氮(nitric oxide,NO)含量,Griess化学比色法测定血清中一氧化氮合酶(nitric ox-ide sythase,NOS)活性的变化。结果与正常组相比,模型组大鼠心功能抑制,RK/LK和LVM/BW比值升高,心脏、肾脏和腹主动脉IMD的水平及血浆中IMD和AngⅡ的含量升高,血清中NO和NOS的含量降低;与模型组相比,大蓟组大鼠心功能明显增强,RK/LK和LVM/BW比值及血浆中AngⅡ的含量降低,心脏、肾脏和腹主动脉IMD的水平升高,血浆中IMD的含量、血清中NO和NOS的含量升高,差异均有显著性。结论大蓟通过增加IMD的含量激活NO/NOS对肾性高血压有降压作用。ObjectiveTo approach the effect of Daji alcohol extract on the change of intermedin（IMP） in two kidney-one clip hypertensive rats.MethodsThe renal hypertensive model in rats was established by two kidney-one clip.Heart function,the ratio of left ventricular mass to body weight（LVM/BW）,and the ratio of right kidney weight to left kidney（RK/LK） were tested.The IMD level of heart,kidney and abdominal aorta were tested by immunohistochemistry method.Radiation immunological method was used to detect plasma levels of IMD and angiotensinⅡ（AngⅡ）.The content of nitric oxide（NO）in serum were tested by nitrate reductase method,and the activity of nitric oxide synthase（NOS） in serum was tested by Griess chemistry colorimetric technique.ResultsCompared with normal group the heart function was inhibited,RK/LK and LVM/BM were elevated,ZMD level in heart,kidney,abdominal aorta and ZMD and Ang Ⅱ contents in serum were increased,NO and NOS in serum were reduced in model group.ConclusionDaji can decrease blood pressure by increasing the content of IMD and activating NO/NOS in renal hypertension.
