摘 要:目的观察艾灸对糖尿病(DM)性阴茎勃起功能障碍(ED)大鼠阴茎勃起组织细胞凋亡情况的影响。方法以链脲佐菌素(STZ)诱导建立DM大鼠模型后再用阿朴吗啡(APO)阴茎勃起实验筛选DM性ED大鼠。以正常大鼠作为空白组,并将发生ED者随机分为模型组和模灸组。模灸组给予"肾俞""三阴交"麦粒灸治疗2个疗程后,处死6组大鼠,取其阴茎勃起组织,采用原位末端标记法(TUNEL)检测细胞凋亡情况,免疫组化法检测凋亡相关基因Bax和Bc l-2的表达情况。结果 模灸组大鼠阴茎勃起组织细胞凋亡指数和Bax基因的表达均较模型组明显下降(P<0.01),而其Bc l-2基因的表达却较模型组明显升高(P<0.01),同时,其Bc l-2/Bax的值较模型组增高且比值大于1。结论 艾灸能改善DM性ED大鼠阴茎勃起组织的细胞凋亡情况,其作用机制可能是通过减少DM性ED大鼠阴茎勃起组织中Bax基因的表达,增加Bc l-2基因的表达,并提高Bc l-2与Bax的比值来实现的。Objective To investigate the effect of moxibustion of apoptosis in penis cavemous tissue of rats with diabetic ED.Methods Male Sprague-Dawley rats with normal erectile function were injected with streptozotozin(STZ) to establish experimental model of Diabetes Mellitus(DM).Then apomorphine(APO) was injected into all of DM rats to select ED rat model with DM.The normal rats were in blank group and the ED rats were randomly divided into model group and moxibustion group.The model group was not treated while the model-moxibustion group was treated with moxibustion on acupoints "Shenshu"(BL23) and "Sanyinjiao"(SP6) by two courses of treatment.All rats were killed and their penis cavernous tissues were collected.The apoptosis was evaluated by means of terminal deoxynucleotidyl transferase mediated d-UTP nick end labeling(TUNEL).Immunohistochemistry was employed to detect the protein of Bax and Bcl-2.Results The apoptotic index of penis cavernous tissues and the average optical demsity of Bax gene in model-moxibustion group was lower than model group(P0.01),while the average optical demsity of Bcl-2 gene and the ratio between Bcl-2 gene and Bax gene in model-moxibustion group was higher than those of model group(P0.01),and Bcl-2/Bax1.Conclusion Moxibustion can improve the apoptotic state in penis cavernous tissues of rats with diabetic ED,and the mechanisms may be related to the decrease of Bax gene expression,the increase of Bcl-2 gene expression,and the increase of the ratio between Bcl-2 gene and Bax gene in penis cavernous tissues of rats with diabetic ED.
