摘 要:目的探讨HLA-G基因单核苷酸多态性(SNP)与广西儿童支气管哮喘(哮喘)发病的关联性。方法对广西哮喘患儿88例(哮喘组)及112例健康对照儿童(健康对照组)采用基于基质辅助激光解吸电离-飞行时间质谱技术检测了HLA-G基因SNP的分型变化(SNP位点包括rs1736936、rs3115630、rs1632947、rs1233334、rs1736933、rs1736932、rs1631950、rs1704和rs1063320),探讨HLA-G基因SNP与哮喘发生的关联。采用SPSS 13.0软件进行分析。结果发现rs1063320、rs1632947共2个位点的基因型构成比在哮喘组与对照组有显著性差异(P<0.05);rs1631950、rs1736932、rs1736933、rs1736936共4个位点的基因型在哮喘组和健康对照组有临界性差异(P=0.05~0.06)。其中rs1063320位点的CC基因型携带者与GG基因型携带者相比,哮喘危险性增加了1.27倍(P<0.05)。rs1632947位点的TT基因型携带者与CC基因型携带者相比,哮喘危险性增加了1.59倍(P=0.02)。结论 HLA-G基因SNP与广西儿童哮喘的发病相关。rs1063320位点的CC基因型和rs1632947位点的TT基因型是儿童哮喘的易感基因型。Objective To investigate the correlation between HLA - G gene single nucleotide polymorphisms (SNP) and bronchial asthma occurrence in children in Guangxi Region. Methods Totally 88 asthma children (asthma group) and 112 healthy controls (healthy control group) in Guangxi region were included in the study. The HLA - G gene SNP genotypes, including rs1736936, rs3115630, rs1632947, rs1233334, rs1736933, rs1736932, rs1631950, rs1704 and rs1063320 ,were performed by means of matrix assisted laser desorption ionisation - time of flight mass spectrometry method according to the manufacturers instructions. The data were analyzed by SPSS 15.0 software. Results There was a significant difference in the distribution of the genotypes of 2 SNPs ( rs1063320 and rs1632947 ) between asthmatic children and healthy controls ( P 〈 0.05 ), while criticality difference ( P = 0.05 - 0.06 ) was showed in other 4 SNPs ( rs1631950, rs1736932, rs1736933 and rs1736936). The individuals with rs1063320 CC genotype had a 1.27 fold increased risk for developing asthma, compared with GG geno- type ( P 〈 0.05 ). And the risk of children with rs1632947 T/T genotype for asthma was 1.59 fold higher than CC genotype ( P = 0.02 ). Conlusions HLA - G SNPs has correlations with etiology of asthma. Having the genotype of rs1063320 C/C and rs1632947 T/T in the HLA - G are in susceptibility to children asthma.
