肾炎四味片及洛汀新对糖尿病大鼠肾脏足细胞数目的影响

摘　　要：目的探讨肾炎四味片和洛汀新对糖尿病大鼠肾脏足细胞数目的影响。方法建立STZ诱导的糖尿病大鼠模型,将35只大鼠随机分为正常对照组、糖悄病组、肾炎四味片干预组、洛汀新干预组及肾炎四味片与洛汀新联合干预组(联合干预组)各7只。应用WT-1标记足细胞核,观察8周末5组血糖、尿蛋白、血肌酐、肾质量/体质量;光镜及电镜观察肾组织形态学变化,免疫组化检测足细胞数目的变化。结果与正常对照组比较,糖尿病组大鼠血糖、尿蛋白、肌酐清除率、肾质量/体质量明显增加,足细胞数目明显减少(P<0.01);与糖尿病组相比,除血糖外,肾炎四味片干预组、洛汀新干预组及联合干预组上述指标明显改善(P<0.05),而3组间差异无统计学意义(P>0.05)。足细胞数目与尿蛋白呈负相关(r=-0.95,P=0.0036)。结论肾炎四味片及洛汀新具有改善糖尿病肾病发展过程中足细胞数目减少、减轻尿蛋白的作用。Objective To investigate the effect of Shenyansiwei tablets and benzapril on the podocyte number in renal tissue of rats with diabetic nephropathy.Methods STZ-induced diabetic rats were established,then these rats were assigned randomly into 5 groups,normal control group,diabetic group,Shenyansiwei tablets group,benzapril group and combine Shenyansiwei Tablets and benzapril group.Used the WT-1 to stain podocyte nucleus,at the 8th week,24 h urine samples were collected for analysis blood sugar,proteinuria,and creatinine.Then rats were sacrificed,serum creatinine,renal pathological change,podocyte number were observed.Results At the 8th week,compared with normal control group,blood sugar, proteinuria,the clearance of creatinine were increased dramatically in the diabetic group,Shenyansiwei tablets group,benzapril group and combined group,and the podocyte number decreased dramatically（P0.01 ）.Compared with diabetic group, except blood sugar,the proteinuria and the clearance of creatinine normalized dramatically in Shenyansiwei tablets group, benzapril group and combined group（P0.05）,and there were no significant difference of these data among these three treated group（P0.05）.The podocyte number correlated negatively with proteinuria（r = -0.95,P =0.0036）.Conclusion Shenyansiwei tablets and benzapril could decrease proteinuria and podocyte number and improve renal function.
