摘 要:目的:考察加味佛手散对大鼠子宫内膜异位症的治疗作用及其在免疫学方面的机制。方法:自体移植法建立大鼠子宫内膜异位症模型,灌胃加味佛手散28 d后,用游标卡尺检测移植物体积,电子天平称量脾脏质量,免疫组化法检测移植物中白介素(IL)-8的表达,ELISA法检测血清及腹腔液中肿瘤坏死因子(TNF)-α的表达,Western blot法检测异位组织中核转录因子-κB亚基p65(NF-κB p65)及NF-κB抑制蛋白IκBα的表达。结果:加味佛手散0.045,0.09,0.18 g.kg-1能明显减小大鼠子宫内膜异位组织的体积,加味佛手散0.18 g.kg-1能显著增加脾脏指数,加味佛手散0.09,0.18 g.kg-1能够降低IL-8在异位组织中的表达,显著降低血液及腹腔液中TNF-α含量,并能降低异位组织中NF-κB p65含量及增加IκBα的含量。结论:加味佛手散可显著抑制大鼠子宫内膜异位症移植物生长,其作用机制可能与改善机体免疫环境有关。Objective: To explore the effect and mechanism of Jiawei Foshou San on the accretion ectopic endometrium of rats. Method: Endometriosis model was established by surgical implant of endometrial tissue which belongs to its body in rats. Jiawei Foshon San was administrated to the model rats. Twenty-eight days later, the length of ectopic endometrium was measured by vernier caliper, the spleen was weighed by electronic balance, the content of tumor necrosis factor (TNF) -a in blood serum and peritoneal fluid was detected by ELISA test, the expression of interleukin (IL)-8 in ectopic endometrium was detected by immunohistochemical method, the expression of NF-κB p65 protein and inhibitory. κBα (IκBα) protein in ectopic endometrium were analyzed by western blot. Result: Jiawei Foshou San 0. 045, 0.09, 0. 18 g · kg^-1 group reduced the volume of ectopic endometrium. Jiawei Foshou San 0.18 g · kg^-1 group raised the spleen exponent of model rats. Jiawei Foshou San 0.09,0.18 g · kg^-1 group decreased the content of TNF-α in blood serum and peritoneal fluid, and the content of IL-8 in ectopic endometrium was also decreased. Jiawei Foshou San can decrease the expression of NF-κB p65 and increase the expression of IκBα in ectopic endometrium. Conclusion: Jiawei Foshou San can inhibit the accretion of endometriosis implants of rats, and its mechanism might be associated with improving the environment of body.
