脑胶质瘤细胞SHG44与U87-MG辐射敏感性差异及其机制初步研究

摘  要:目的本实验通过研究脑胶质瘤细胞SHG44与U87-MG的辐射敏感性及COX-2 mRNA表达水平差异,初步探讨胶质瘤细胞辐射敏感性差异的可能机制,为临床有效治疗胶质瘤提供实验依据。方法细胞克隆法检测不同60Co-γ射线照射剂量下细胞克隆存活率;RT-PCR法检测两种细胞COX-2 mRNA表达水平。结果实验表明两种胶质瘤细胞的辐射敏感性存在差异,U87-MG的辐射敏感性高于SHG44细胞,其差异具有统计学意义(P<0.05);两种细胞的COX-2 mRNA表达水平不同,U87-MG细胞COX-2 mRNA表达水平高于SHG44细胞,其差异具有统计学意义(P<0.01)。结论本实验研究认为脑胶质瘤U87-MG的辐射敏感感性高于SHG44细胞,其机制与其COX-2 mRNA表达水平密切相关。Objective To study the radiosensitivity and COX-2 mRNA expression differences of glioma cells SHG44 and U87-MG,and preliminary investigate the possible mechanism of radiosensitivity differences for different glioma cells,so as to provide some experiment support for clinical treatment of glioma.Methods Colony Formation Assay was used to investigate the effect of 60Co γ-irradiation on cell colony formation rate.Reverse Transcription-PCR assay were used to investigate the levels of COX-2 mRNA expression on glioma cells(SHG44 and U87-MG).Results Both the radiosensitivity(P0.05) and COX-2 mRNA expression(P0.01) of U87-MG is signif icant higher than that of SHG44.Conclusion The radiosensitivity of U87-MG is higher than SHG44 has a close relation with its COX-2 mRNA expression.
