摘 要:目的探讨中药方剂四君子汤对烟曲霉菌合成毒性代谢产物的抑制作用。方法不同剂量四君子汤处理的5×106烟曲霉菌孢子用DMEM液培养48 h,收集不同组的培养液滤液,计算其不同组培养液滤液作用的HeLa细胞和红细胞生存率;比较抑制50%细胞生长的不同组培养液滤液的稀释度;观察100μl 0.3%H2O2在沙氏固体培养板上对一定剂量四君子汤处理的烟曲霉菌孢子的抑菌圈大小。结果四君子汤处理的烟曲霉菌孢子培养液滤液作用的HeLa细胞和红细胞生存率显著高于无药物处理组(P<0.05),抑制50%细胞的药物组孢子培养液滤液的稀释度低于无药物组,结果均呈现剂量-效应关系;100μl 0.3%H2O2在沙氏固体培养板上作用一定剂量四君子汤处理的烟曲霉菌孢子所产生抑菌圈直径显著大于无药物处理组(P<0.05),结果呈现剂量-效应关系。结论中药方剂四君子汤可以抑制烟曲霉菌培养时毒性代谢产物的产生,治疗烟曲霉菌所致的肺部感染具有潜在的应用价值。Objective To explore the inhibitory effect of Si-jun-zi decoction on the production of toxic metabolites of Aspergillus fujmigatus.Methods 5×106 conidia of Aspergillus fumigatus treated with 100,200,300 μl and without Si-jun-zi decoction were cultivated in DMEM medium for 48 h.The culture medium of conidia in four groups was collected and filtrated.The survival rate of HeLa cells and chicken red blood cells treated with filtrated culture medium from four groups were statistically compared and the dilution of filtrated medium to inhibit 50% cell growth was measured.Inhibition zone diameter was measured on Sabouraud dextrose agar plate on which 100 μl 0.3% H2O2 interacted with conidia.Results The survival rate of HeLa cells and chicken red blood cells in Si-jun-zi decoction groups were significantly higher than those without Si-jun-zi decoction group((P0.05).The dilution of filtrated medium of conidia in Si-jun-zi decoction groups to inhibit 50% cell growth was lower than that in non Si-jun-zi decoction group.The activity of Si-jun-zi decoction to decrease the toxin production in filtrated medium of conidia was dose-dependent.The inhibition zone diameter was bigger in Si-jun-zi decoction groups than in non Si-jun-zi decoction group.The effect of Si-jun-zi decoction to enhance the sensitivity of conidia to H2O2 was dose suggesting dependent.Conclusion Si-jun-zi decoction can inhibit the production of toxic metabolites by aspergillus fumigatus significantly,showing that Si-jun-zi decoction has potential therapeutic value against lung infection by aspergillus fumigatus.
