摘  要:目的研究朱砂及其复方朱砂安神丸对小鼠肝脏的急性毒性,并在此基础上探讨其对肝脏细胞色素P450酶基因表达的影响。方法小鼠随机分为空白对照组,朱砂安神丸高剂量组(20 g·kg-1ig)、朱砂安神丸低剂量组(10 g·kg-1ig)和朱砂组(0.3 g·kg-1ig),各组小鼠均早晨给药1次,8 h后断头处死小鼠,分别检测肝中汞蓄积量,肝功能,肝指数,并应用实时荧光定量PCR(RT-PCR)的方法检测其对细胞色素P450酶基因表达的影响。结果与空白对照组相比,朱砂和朱砂安神丸组小鼠肝脏ALT和AST均有所增高,但差异无统计学意义;给药三组小鼠肝脏中汞蓄积量均有所增高,并呈现一定的剂量相关性,但无统计学意义;在对基因表达的影响方面,单次给药后,与空白对照组相比,朱砂组Cyp2 a4、Cyp2b9、Cyp 2 f2、Cyp8b的基因表达下降,Cyp3 a11和Cyp3 a44的基因表达增高:朱砂安神丸低剂量组Cyp2b9和Cyp2 f2的基因表达下降;各组对其他基因的表达情况无统计学意义。结论单次给药后,汞在小鼠肝脏中蓄积很少,对肝脏未造成明显损伤,但朱砂和朱砂安神丸对细胞色素P450酶的基因表达具有一定的诱导和抑制作用。Objective To study the effects of Cinnabar and Zhusha Anshen Pill on the expression of cytochrome P450 mRNA in mouse liver.Methods Mouse were orally given Zhusha Anshen Pill(10,20 g·kg-1) and Cinnabar(0.3 g·kg-1),8 h later,the concentration of mercury,liver index,and mRNA levels of cytochrome P450 were determined.Results Zhusha Anshen Pill had no effect on ALT and AST,and no significant accumulation of mercury in mouse liver was observed.The RT-PCR analysis indicated that the expression of Cyp2a4,Cyp2b9,Cyp2f2 and Cyp8b was decreased in cinnabar group,but the expression of Cyp3a11 and Cyp3a44 was increased.The expression of Cyp2b9 and Cyp2f2 was decreased in the low dose Zhusha Anshen Pill group.Conclusion Cinnabar and Zhusha Anshen Pill can affect the expression of cytochrome P450 mRNA,but has no obvious liver injury.
