摘 要:目的观察蒙药复方白脉散对大鼠脑缺血再灌注神经损伤的保护作用。方法将健康成年大鼠随机分为3组:假手术组、缺血再灌注模型组、白脉散组。术前灌胃给药7 d,末次给药30 min后,采用改良的线栓法制作大鼠大脑中动脉闭塞再灌注(MCAO/R)模型。苏醒后对其神经功能进行评分;缺血2 h再灌注24 h后观察病灶处病理学变化,测定脑匀浆上清超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、还原型谷胱甘肽含量、一氧化氮合酶(NOS)活性等。结果与模型组比较,蒙药复方白脉散组病灶处皮层神经元肿胀坏死有所减轻;脑匀浆上清SOD活性和GSH含量明显升高,MDA含量显著降低(P<0.05);T-NOSi、NOSc、NOS活性均明显降低(P<0.05)。结论蒙药复方白脉散对大鼠局灶性脑缺血有明显的保护作用,其机制可能是通过有效的拮抗自由基损伤,提高脑组织抗氧化能力来实现的。Objective To investigate the neuroprotective effects of Mondlian medicine BaiMai Power on cerebral ischemia-reperfusion injury in rats. Methods 36 Wistar rats were divided into three groups randomly:sham-operation group,cerebral ischemia-reperfusion injury group,Mongolian medicine BaiMai Power treatment group.Intraluminal thread method was applied to establish the middle cerebral artery occlusion reperfusion model(MCAO/R)in rats.The rats of Mongolian medicine BaiMai Power group were given Mongolian medicine BaiMai Power(200 mg/kg) by oral gavage once a day.After 24 h ischemia-reperfusion,the pathological changes were observed in lesions,and the activities of superoxide dismutase(SOD) and nitric oxide synthase(NOS) and the contents of malondialdehyde(MDA) and glutathione(GSH) in brain were determined. Results Compared with model group,the contents swelling and necrosis of cortical neurons in Lesions were lessened in Mongolian medicine BaiMai Power treatment group.GSH content and SOD activity significantly increased(P0.05),and MDA content and NOS activity(which included these three subtypes T-NOS,cNOS iNOS)significantly decreased(P0.05). Conclusion Mongolian medicine BaiMai Power can redue the pathological changes of cerebral-ischemia rats,and the mechamism may be related with the improverment of antioxidant capacity of brain tissue and the decrease of oxidative injury.
