摘　　要：目的观察羟基积雪草苷(MC)对大鼠急性脊髓损伤(SCI)的保护作用。方法制备SCI模型,成年大鼠150只随机分为假手术组、模型组、MC高、低剂量治疗组及甲基强的松龙(MP)对照组,每组30只,术后24 h7、2 h、7 d1、4 d2、8 d各个时间点观察大鼠后肢运动功能及脊髓病理变化情况,检测脊髓中丙二醛(MDA)、神经元特异性烯醇化酶(NSE)含量及超氧化物歧化酶(SOD)活性。结果高、低剂量MC及MP用药能明显提高SCI大鼠行为学评分,改善病理损伤,降低MDA含量,提高SOD活性,增加NSE表达,MC的作用呈剂量依赖性。结论 MC可减轻SCI大鼠的脊髓损伤,此作用可能与抑制损伤脊髓的脂质过氧化反应有关。Objective To observe the effect of madecassoside（MC） on the acute spinal cord injury（SCI） in rats. Methods SD rat model by modified Allens assay for SCI was adopted.150 rats were divided randomly into five groups： sham-operative group,model group,and the groups treated with madecassoside（60mg/kg and 20mg/kg）and methylprednisolon（30mg/kg）.At 24h,72h,7d,14d,28d after operation,motor function of the rat hind limb was determined by the BBB（Basso Beattie and Bresnahan）,pathological state of spinal was observed by HE,and the activity of SOD and the contents of MDA and NSE were detected. Results Compared with the sham-operative group,at each time point the motor function of the rat hind limbs in acute spinal cord injury model group was decreased,the level of MDA was increased,and the activity of SOD and level of NSE were decreased in spinal cord tissue;treatments with MC（60 and 20mg/kg） and MP（30mg/kg） could significantly improve the motor function of the rat hind limbs and pathological outcomes,markedly decrease the elevation of MDA level and increase the activity of SOD and the content of NSE,and the effect of MC was in dose-dependent manner. Conclusion MC has protective effect on SCI and the mechanism may be related with inhibiting lipid peroxidation and decreasing the level of free radical.
