摘  要:目的探讨补骨脂汤对血管痴呆海马内神经元的保护作用及生长相关蛋白-43基因表达(GAP-43)基因表达的影响。方法将60只SD大鼠分成假手术组、模型组、补骨脂汤低剂量组(以下简称补低组)、补骨脂汤高剂量组(以下简称补低组)及银杏叶片组,每组12只,制备血管性痴呆大鼠模型,手术后第3天开始给药,时间1个月,采用HE染色及RT-PCR的方法,分析海马CA1区的神经元的数量和mRNA的表达量。结果补高組、补低组大鼠海马神经元的数量明显高于模型组(P<0.05),GAP-43基因表达的mRNA量明显高于模型组(P<0.05)。结论补骨脂汤可能通过提高GAP-43基因的表达水平,使血管性痴呆海马内的神经元得到很好的保护。ObjectiveTo investigate the effect of Buguzhi decoction on hippocampal neurons of vascular dementia model rats. MethodsSixty rats with vascular dementia were randomly divided into five groups:control group,sham-operation group,model group,Buguzhi decoction groups and Folium Ginkgo group.The rat model with VaD was established by bilateral occlusion of both common carotid arteries(2-V0).The changes of neurons were detected by HE staining.The expression of GAP-43 gene was evaluated by RT-PCR. ResultsLight microscope showed that the number of neurons in hippocampal CA1 field in Buguzhi decoction was higher than model group.Buguzhi decoction could significantly increase mRNA expression of GAP-43 gene in hippocampus of vascular dementia model rats,compared with model group(P0.05). ConclusionBuguzhi decoction has protective effect on neurons of vascular dementia model rats by increasing the expression of GAP-43 gene.
