摘  要:目的构建巨噬细胞炎症模型,比较研究半枝莲黄酮、洛伐他丁及野黄芩苷对细胞因子IL-1β、IL-6和TNF-α基因表达情况及含量的影响。方法培养巨噬细胞RAW264.7,利用脂多糖构建细胞炎症模型,并在不同时间收集细胞或细胞上清液,通过RT-PCR与ELISA法分别检测基因的表达情况及含量变化。结果半枝莲黄酮可以有效降低IL-6和TNF-α基因的表达,并且能够降低IL-6和TNF-α的含量,其中以IL-6最为显著,其抗炎作用要远远好于单体药物。结论半枝莲黄酮可以通过有效降低细胞因子基因的表达及其含量来挥发其抗炎作用。ObjectiveTo observe the effects of the total flavones from Scutellaria barbata D.Don,lovastatin and scutellarin on gene expression and contents of cytokine IL-1β,IL-6 and TNF-α in LPS-induced inflammatory macrophage model. MethodsInflammatory model was constructed with LPS through cell culture with macrophage RAW 264.7.Cells and supernatant were collected at different times.Gene expression and cytokine concentrations were detected with the methods of RT-PCR and ELISA. ResultsCompared with monomer lovastatin and scutellarin,the total flavones from Sculellaria barbata D.Don could effectively decrease gene expression and cytokine concentrations of IL-6 and TNF-α,especially for IL-6. ConclusionThe total flavones from Scutellaria barbata D.Don can effectively exert its anti-inflammatory effects through decreasing gene expression and contents of cytokines.
