摘  要:目的探讨小补心汤总黄酮(XBXT-2)含药血清的神经保护作用。方法采用血清药理学方法分别制备大鼠的空白血清、氟西汀(10 mg/kg)和XBXT-2(25,50,100,200 mg/kg)含药血清。采用皮质酮损伤大鼠肾上腺嗜铬细胞瘤细胞(PC12)、人神经母细胞瘤细胞(SH-SY5Y)及原代培养大鼠海马神经元为体外药理学模型,应用ATP生物发光法检测XBXT-2及氟西汀含药血清对损伤细胞存活率的影响。结果皮质酮(50~300μmol/L)能够浓度依赖性地抑制SH-SY5Y的存活,氟西汀与XBXT-2(25,50 mg/kg)含药血清能显著提高皮质酮(100μmol/L)损伤的SH-SY5Y的存活率(P<0.01,P<0.05,P<0.01);与空白对照组比较,200μmol/L皮质酮导致PC12的存活率显著下降(P<0.01),氟西汀与XBXT-2(25,50,100 mg/kg)含药血清能显著对抗这种损伤(P<0.05,P<0.01,P<0.05);100μmol/L皮质酮使原代海马神经元存活率显著下降(P<0.001),XBXT-2(25、50 mg/kg)对这种损伤具有显著的保护作用(P<0.05,P<0.001),其作用与阳性药氟西汀含药血清相当。结论 XBXT-2含药血清对皮质酮所致的神经细胞损伤具有明显保护作用,这可能是其抗抑郁效应的重要细胞机制之一。ObjectiveTo investigate the neuroprotective effect of serum containing the total flavonoids extracted from Xiaobuxin-Tang(XBXT-2).MethodsSerum pharmacology experiments were used to obtain rat serum including control serum,and serum containing fluoxetine(Flu,10 mg/kg) or XBXT-2(25,50,100 and 200 mg/kg),respectively.Cultured PC12 cells,SH-SY5Y cells and primary culture of hippocampal neurons from neonatal rats were incubated with corticosterone(Cort) for 48 h in the presence of control serum or serum containing fluoxetine or XBXT-2.The cell viability was measured by ATP-luminescent cell viability assay.ResultsAfter incubation with Cort(50-300 μmol/L),the cell viability of SH-SY5Y significantly decreased in a dose-dependent manner.Flu and XBXT-2(25 and 50 mg/kg) serum could significantly increase the cell viability of SH-SY5Y cells with Cort(100 μmol/L) co-incubation(P0.01,P0.05 and P0.01).Compared with the control group,the cell viability of PC12 exposure to Cort(200 μmol/L) was significantly decreased(P0.01),while the injury was significantly eased by serum containing Flu and XBXT-2(25,50 and 100 mg/kg)(P0.05,P0.01 and P0.05).XBXT-2(25 and 50 mg/kg) could protect primary culture of hippocampal neurons from the lesion induced by Cort(100 μmol/L)(P0.05 and P0.001),and the positive control fluoxetine(10 mg/kg) serum exerted the same effect.ConclusionSerum containing XBXT-2 shows significant protection against nerve injury induced by Cort,which may be one of the important cellular mechanisms underlying the antidepressant action of XBXT-2.
